Kudzu vine is widely distributed in world and has abundant resources.As a by-product of pueraria lobata,kudzu vine is usually neglected by people and abandoned or burn,which resulted in waste of plant resources and environmental pollution.A variety of bioactive ingredients in kudzu vine were found and known after many years of studies.Flavonoids is the main ingredient and plays a positive action in anti-tumor,anti-virus,immune function,lipid-lowering and other pharmacological activity.In this experiment,different doses of kudzu vine flavonoids extract were added in the diet of broilers to determin the effects of kudzu vine flavonoids extract on the growth performance and lipid metabolism of broilers.At the same moment,the mechanism of kudzu vine flavonoids extract were further explored to study lipid metabolism according to the express level of blood lipid,endocrine hormone,the activities and gene expression of lipid metabolism-related enzymes.In the experiment,240 one-day-old AA broilers were randomly divided into 4 groups,6 replicates in each group and 10 broilers replicates.These groups were set in the control group(basal diet),0.07%,0.14%,0.21% kudzu vine flavonoids extract groups(basal diet + 0.07%,0.14%,0.21% kudzu vine flavonoids extract),the experiment lasted 42 days.The production performance,immnunty organ index,body fat,serum lipid contents,endocrine hormone,the activities and gene expression of lipid metabolism-related enzymes were detected and analysed in this experiment.The results showed that:(1)Kudzu vine flavonoids extract in the diet didn't cause significant changes of the production performance,chest muscle rate and leg muscle rate of broilers(P>0.05).Kudzu vine flavonoids extract in the diet increased remarkably immune organ indexes.0.21% of kudzu vine flavonoids extract improved significantly fabricius index of 21 days-old broiler(P<0.05),and significantly improve the chicken liver index and spleen index of 42days-old broilers(P<0.05).(2)At 21 days-old,0.07% of kudzu vine flavonoids extract had significantly reduced the abdominal fat percentage and sebum rate(P<0.05),but had no significant effect on fat content of the breast muscle and the leg muscle(P>0.05).At 42 days-old,abdominal fat rate significantly reduced in 0.14% group and 0.21% group than the control group(P<0.05),but the fat content of the breast muscle didn't had a significant change in 0.21%group than in the control group(P>0.05).Take the above considered,individually adding0.07% at early stage and 0.21% at later stage of kudzu vine flavonoids extract was very helpful for broilers to maintain a better body fat and muscle fat content.(3)Kudzu vine flavonoids extract could decrease the contents of TC,TG and LDL-C,and increase the content of HDL-C in broilers.Kudzu vine flavonoids extract increasedserum Apo-AI,Apo-AII concentration of 21 days-old broilers,and even 0.14% increased significantly(P<0.05).Kudzu vine flavonoids extract reduced the concentration of Apo-CIII(P>0.05).At 42 days-old,kudzu vine flavonoids extract reduced serum Apo-AI,Apo-AII and Apo-CIII concentration of broilers.Compared with the control group,0.14%group and 0.21% group decreased significantly in serum Apo-AI concentration of broilers(P<0.05),and 0.21% group decreased significantly in serum Apo-CIII concentration of broilers(P<0.05).(4)Kudzu vine flavonoids extract could promote the express of insulin,glucagon and thyroxine.0.14% kudzu vine flavonoids extract increased significantly the insulin concentration of 21 days-old broilers(P<0.05),0.21% kudzu vine flavonoids extract increased significantly the concentrations of glucagon(P<0.05).Kudzu vine flavonoids extract inhibited remarkably the activity of fat-related enzymes FAS and ACC,and enhanced HSL activity of 42-days-old broilers(P<0.05).Therefore,kudzu vine flavonoids extract could activate the PPAR? pathway and promote the mRNA expression of CPT-1?,ACO and L-FABP,which resulted in ?-oxidation of fatty acid and lipolysis.0.21% kudzu vine flavonoids extract inhibited fat synthesis through reducing the mRNA expression of FAS,ACC.In summary,kudzu vine flavonoids extract didn't change the growth performance of broilers,but it could inhibit fat synthesis and enhance lipolysis,which resulted in reduction of the rate of abdominal fat and sebum in broilers.Thus,kudzu vine flavonoids extract played a positive role in body fat deposition of broilers,especially 0.21%.
