| Wheat gibberellic disease(Fusarium head blight,FHB)is a kind of important diseases on cereal crops,and were spread in almost all of the world’s wheat production areas,especially seriously harmed the mild and humid and semi-humid regions.FHB has been distributed in majority of the most important wheat diseases.In our country,there are mainly Fusarium asiaticum and Fusarium asiaticum,and Fusarium asiaticum accounted for the vast majority of Fusarium asiaticum.Benzimidazole fungicide control FHB for 30 years,and cause a serious resistance problem Jits application scope and prospects are constrained by the very serious.Phenamacril(test number:JS399-19)is compound by the national center of southern pesticide in jiangsu in 1998,it is better highly reactive compounds to Fusarium asiaticum,and can strongly inhibit fungus hypha growth.Through whole genome research of YP-1,Zhitian discovered the myosin heavy 5(myosin5)point mutation in myosin-5 give rise to resistance to the fungicide phenamacril in Fusarium asiaticum.,and different point mutations lead to different levels of resistance.In Fusarium asiaticum,there are three kinds of myosin,respectively is the class Ⅰmyosin5,class Ⅱ myosin2 and class V myosin2B,and knock out or myosin5 myosin2B will cause death of Fusarium asiaticum.In Fusarium asiaticum.there are three myosin genes,myosin 5(myosin5)point mutation led to resistance to fungicide phenamacril in Fusarium asiaticum,then whether the other two myosin genes play a role in regulation resistance level to the fungicide phenamacril in Fusarium asiaticum is also very necessary.In yeast,protein Smyl collaborated with myosin2B,through the yeast two hybrid method experiment,we proved FaSmyl and FaMyo2B has strong interaction relations in Fusarium asiaticum.For a detailed functional analysis of FaMyo2B,FaMyo2and FaSmyl,we generated targeted disruption or deletion mutants by transformation of the gene replacement cassette HPH-HSV-tk in a phenamacril-sensitive strain(2021)and in a phenamacril-resistant strain(Y2021A)of F.asiaticum,we obtained FaMyo2B disruption mutants,FaMyo2 and FaSmyl deletion mutant in Wild strain 2021 and high resistance strain to phenamacril.Here,we determined whether there is a functional link between phenamacril resistance and the myosin proteins FaMyo2,FaMyo2B and FaSmyl in Fusarium asiaticum,which is an important factor for Fusarium head blight of wheat.The class V myosin motor protein Myo2 plays an important role in in budding yeast and participates in numerous membrane trafficking events.We found that FaMyo2B acts jointly with FaSmyl to affect resistance to phenamacril in F.asiaticum.We also found that FaMyo2B disruption mutant,FaMyo2 and FaSmyl deletion mutants were defective in hyphal branching,conidiation,and sexual reproduction.OFaMyo2 also resulted in the disordered construction of nuclei and septa and an increased sensitivity to substance that undermined the cell wall.In addition,the FaMyo2,FaMyo2B and FaSmyl mutants had weaken the pathogenicity on flowering wheat heads and wheat coleoptiles.Taken together,the laboratory findings confirmed that FaMyo2,FaMyo2B and FaSmyl are required for several F.asiaticum developmental processes and activities,which help us better understand the resistance mechanism and find the most effective approach to control FHB. |
