Pig is a typical single stomach animal with a developed colon,which contains a large number of microorganisms that can use fiber.Suhuai pig has the characteristics of resistance to coarse feeding,fast growthing and high fecundity,which contains 25%of Huai pig blood.Sooner or later of sow sexual maturity and the group sexual maturation rate are very important in breeding industry,and the reproductive ability of the sow is directly determined by the development of follicles.Dietary fiber is essential for pigs in normal physiological metabolism and plays an important role in pig production.The appropriate quantity and content of the fiber can improve the production performance and reproductive performance of pig.Besides,adding some fiber in the diet can save grain and alleviate the contradiction between humans and animals,which is conducive to the sustainable development of animal husbandry in China.Thus,the dietary fiber has become a research focus on the reproductive performance of sows in recent years.In the current study,Suhuai gilts were fed with different content of dietary fiber(the control group contained 5.41%dietary fiber,the experimental group contained 7.58%dietary fiber)to study the effects of dietary fiber on sex maturation and follicle development.The results are as followed.1.Effects of dietary fiber on body weight and sex maturition in the giltsAfter 75 days of feeding,the results showed that the body weight and the daily weight gain of the experimental group are significantly lower than that of the control group.The number and the rate of sex maturation of the experimental group are significantly lower than that of the control group.2.Effects of dietary fiber on uterus weight and ovary weight in the giltsCompared with the control group,dietary fiber has no significant effects on uterine weight and ovary weight of the experimental group in the same stage(follicular stage,luteal stage and sexual immature stage).In the control group,the uterus weight in the follicular stage and the luteal stage is extremely significant higher than that in the sexual immature stage;the ovarian weight in the follicular stage and the luteal stage is significantly higher than that in the sexual immature stage.In the experimental group,the weight after test in the follicular stage is significantly higher than that in the sexual immature stage.The uterus weight and ovary weight in the follicular stage and the luteal stage are significantly higher than that in the sexual immature stage.This indicated that dietary fiber had some effect on the ovary weight of the experimental group.3.Effects of dietary fiber on follicular development and the number of corpus luteum in the giltsCompared with the control group,dietary fiber has no significant effects in primordial follicles,primary follicles,secondary follicles,antral follicles,atretic follicles and corpus luteum of the experimental group at the same stage(follicular stage,luteal stage and sexual immature stage).No matter in the control group or in the experimental group,there was no significant difference among the follicular stage,the luteal stage and the sexual immature stage in primordial follicles,primary follicles,secondary follicles,antral follicles and atretic follicles.4.Expression of Smad4 on different follicles and corpus luteumImmunohistochemical analysis displayed that Smad4 expressed in different ovarian follicles and corpus luteal.In the primordial follicle and the primary follicle,Smad4 expressed in granulosa cells and interstitial cells;in the secondary follicle,Smad4 expressed strongly in granulosa cells and weakly in interstitial cells;in the antral follicles,Smad4 mainly expressed in granulosa cells;Smad4 also expressed in corpus luteum of the sex maturation gilts.Compared with the control group,the expression of Smad4 in different follicles are the same as the experimental group.In conclusion,the present study initially indicated that high fiber diet has effect on sex maturation and the ovary weight,but has no effect on follicular development.
