Microorganism in grain storage such as bacteria,fungi and actinomyces exist from the period of harvest to the period of storage.Once the storage condition is suitable for microorganism's growth and reproduction,the quality and safety of the stored paddy may be influenced.Fungi are more harmful to stored paddy than bacteria or actinomyces.Fungi can make the stored paddy seriously moldy.What's more,some species of fungi can secrete mycotoxins which are deadly to human and animal.There are different fungi in paddy stored in different places.The number and the species of fungi are constantly changing during the storage.This change is related to the storage conditions,such as the temperature and the humidity.Denatumig Gradient Gel Electrophoresis(DGGE)is a kind of molecular bio-technique and can be used to research microbial diversity and microbial evolution.1.This paper developed a data management system regarding the fungi evolution in stored paddy,and the system has been successfully applied online.The system is based on Browser/Server structure,and uses Spring + MyBatis as the development framework,JSP as server-side development technology,Oracle as DBMS,Tomcat as Web server.The main functions of the system are as following:(1)The system can storage and retrieve the morphology and the cultural characteristics of different fungi,which can be useful when conducting morphological identification about fungi in stored paddy.(2)The system can store and retrieve the fungi's change or evolution during paddy's storage in different storage places,which can be useful to analyze the storage period and the severity of moldy.Thus the grain depot can select right storage conditions to prevent stored paddy from moldy.(3)The system can store and retrieve bands' position of different fungi in a PCR-DGGE gel image,which can be used to detect fungi in stored paddy quickly without being sequenced.2.This paper researched different image enhancement methods to improve the quality of the gel image,and used Canny edge detection and Harris comer detection to recognize bands in a gel image.Besides,this paper developed a gel image processing software with MATLAB GUI,which includes different image enhancement methods and filtering methods.The software can automatically or manually recognize bands in a gel image.Use the software to acquire the band's location and then search the location in the database to quickly detect ftungi in stored paddy.Compared with Quantity One,which is a commercial 1-D gel image processing software,the accuracy of recognition is extremely close.However,due to the different design target and the development platform,the software is inferior to Quantity One in functions and processing speed.
