Abstract/Summary:
The study was conducted to investigate the effect of IMP on production performance breeding hens and offspring broilers, hatching performance of eggs and the regulation effect of IMP on meat quality of offspring broilers.A total of 864 AA breeding hens of 20 weeks age which we selected from the same genetic background with similar weight and the egg production rate were 5%,The hens were randomly divided to two groups(NIMP group:control diet and IMP group:0.5%inosine acid(IMP)diets),with 6 replicates per group and repeated 72 hens, the processing period for 30 days.Artificially fertilized eggs were collected when hen egg production rate reached 50 percent, and reached the peak when the rate of laying hens,1850 eggs were collected from each group within 7 days, and a total of 3700 eggs were collected for hatching. After hatching,3000 offspring broilers of 1 day age from each group were randomly divided into 4 groups,A total 4 groups respectively noted as:(1)NN group(the group of control diet breeding hen’s offspring broiler with control diet),(2)NI group(the group of control diet breeding hen’s offspring broiler with 0.5%inosine acid diet),(3)IN group(the group of 0.5%inosine acid diet breeding hen’s offspring broiler with control diet),(4)II group(the group of 0.5%inosine acid diet breeding hen’s offspring broiler with 0.5%inosine acid diet),each group of 6 repeat, each repeat 125 broilers, test period 42 days. The study included three trials as follows:The first study was conducted to investigate the effects of exogenous IMP on production performance of breeding hens, egg hatchability performance and offspring broiler growth performance. The results showed that compared with the NIMP group,IMP can improve the production performance of chicken, egg laying rate and qualified rate were higher than the control group increased by 1.53%and 0.41%,but the differences were not significant(P>0.05);IMP significantly improved the egg shape index(P<0.05),significantly reduced the color of egg yolk(P<0.05),extremely significantly reduced the strength of eggshell and eggshell thickness(P<0.01);IMP was significantly advance the of eggs the fertilization rate, hatching rate and hatching rate of IMP group(P<0.05);compared with NN group,IMP had no significant effect on the average daily feed intake(ADFI),the average daily gain(ADG)and the feed conversion ratio(F/G)of 1-42 day(P>0.05),there is no significant effect on breastmuscle ratio and leg muscle ratio(P>0.05);compared with NN group, the abdominal fat rate of II group significantly increased(P<0.05),the lowest in NI group significantly reduce(P<0.05),and IN group was significantly different withⅡ(P<0.05).The second study was conducted to investigate the effects of adding exogenous IMP in diets of breeding hens and offspring broilers on meat quality, energy metabolism and IMP deposition of offspring broilers.Compared with NN group, the results showed that test groups’ breast muscle and leg muscle color redness and yellowness value(b*)had a modest increased, but the difference was not significant(P>0.05);the chest brightness value(L*)of II group decreased significantly(P<0.05);breast and leg muscle 45 min pH value of IN and NI group significantly increased(P<0.05),chest 24 h pH value of IN and NI group significantly increased(P<0.05),24 h pH value of leg muscle was significantly increased(P<0.05);breast and leg muscle of 42 d broilers’ glycogen content, pyruvate kinase(PK)and hexokinase(HK)activity had no significant difference(P>0.05);the content of lactic acid(LD)and lactate dehydrogenase(LDH)activity of breast muscle in IN group and NI group decreased significantly(P<0.05);there had significantly decreased of LD content and LDH activity in II group,IN group and NI group’ leg muscle(P<0.05);Exogenous IMP increased the inosinic acid content of breast muscle and leg muscle 21 d and 42 d of offspring broilers,II group,IN group and NI group compared with NN,21 d group breast muscle were increased by 15.57%,19.67%,and 1.66%,21 d leg muscle were increased by 0.48%,2.88%and 1.44%;42 d breast muscle were enhanced in 10.50%,2.50%and 11.50%,42 d leg muscle respectively increased 0.83%and 2.07%and 9.54%respectively,but the difference was no significant(P>0.05).The third study was conducted to study the effect of adding exogenous IMP in diets of breeding hens and offspring broilers on enzyme activity of IMP metabolism, the gene relative expression of GPAT and ADSL of offspring broilers. The results showed that, compared with NN group,ATP enzyme(ATPase),creatine kinase(CK)and adenosine deaminase(ADA)activity increased to a certain extent, alkaline phosphatase(AKP)and 5’-nucleotidase activity(5’-NT)tendency to reduction in the breast and leg muscles of 21d offspring broilers, but no significant difference between each groups(P>0.05);breast muscle of 42 d offspring broilers significantly increased the activity of CK(P<0.05),pectoral muscle of Ⅱ group and NI group 5’-NT activity significantly decreased(P<0.05);leg muscle’s CK and 5’-NT activity have no significant difference(P>0.05);ATPase and ADA activity increased to a certain extent in breast and leg muscle of 42 d offspring broilers and the activity of AKP certain to reduce, but the differences were not significant(P>0.05);the expression of GPAT gene of 21 d and 42 d offspring broiler breast muscle and leg muscle trended to increase, but the difference was not significant(P>0.05);there is a significant increase of the expression amount of ADSL gene in NI group breast muscle and IN group leg muscle of 21 d broilers;the amount of expression ADSL gene in breast muscle from Ⅱ group and IN group of 42 d broiler chicks increase significantly(P<0.05).In conclusion, the breeding hens diet add inosinic acid can improving hatching performance by reducing yolk color, strength of eggshell and eggshell thickness;exogenous inosine acid can improve the offspring broiler meat quality through reducing breast muscle’s brightness(L*),improve 45min and 24 h pH value and increase offspring broiler inosinic acid deposition which by improve the 42 days offspring broiler CK activity, reduced by 5’-NT activity and increase ADSL gene expression.
