Bemisia tabaci(Gennadius)(Hemiptera:Aleyrodidae)is a species complex composed of at least 36 morphological indistinguishable cryptic species.It has a wide host range.B.tabaci widely distributed in more than 100 countries and regions expect Antarctica.It is one of the most significant and herbivorous pests of agriculture and horticulture in tropical and subtropical,which can damage cottons,vegetables and ornamental plants.It can caused great economic losses.It Transcriptome,it has been widely used in the invasion mechanism,the replacement of cryptic species,the mechanism of drug resistance and the interaction with symbiotic bacteria of B.tabaci.But there are a few studies by using transcriptome to research the relationship between temperature stress and B.tabaci.In our study,we conducted a hazard investigation of B.tabaci in Jiangsu Province and identify the cryptic species of B.tabaci.By using mtDNA C(OI to researched the genetic diversity of the invasion MED.We also used transcriptome to study the relationship between temperature stress and B.tabaci,in order to find the genes and signaling pathways which sensitive to temperature.We paid more attention to the relationship between HSP70 which were found in the transcriptome and temperature stress.The main results were summarized as follows:1.The results of the investigation covering the occurrence degree of B.tabaci in 13 cities of Jiangsu Province in 2016 showed that the occurrence degree of B.tabaci was no serious,and the occurrence degree of B.tabaci was the North>the Center>the South.The results of the analyses on the occurrence degree of five hosts of B.tabaci showed that tomato>cucumber>eggplant>humuls>pumpkin.2.We identified the cryptic species of B.tabaci in Jiangsu Provice from 2015 to 2016.In 2015,we detected 55 locations about 1230 individuals,the result showed that the MED accounted for 86.5%,the MEAM 1 accounted for 13.5%.And all the locations were detected MED,only 16 locatins detected MEAM 1.In 2016,we detected 57 locations about 1748 individuals,the resulted showed that the MED accounted for 91.9%,the MEAM 1 accounted for 8.1%.During all the locations,there were 55 locations found MED,only 9 locations found MEAM 1.The occurrence frequency of MED in 2016 was higher than 2015 and were not found other cryptic soecies in two years.3.We used transcriptional technology checked-10?,26?,43? transcriptional profiles of MED.(1)We found 3024 Unigenes were differentially expressed between 26?-10? including 2087 upregulated Unigenes and 937 downregulated Unigenes.Among these pathways,there were 39 KEGG enrichment pathways(P value<0.05),such Metabolic pathways and Peroxisome.We found 4437 Unigenes were differentially expressed between 26?-43 ?,including 2506 upregulated Unigenes and 1931 downregulated Unigenes.Among these pathways,there were 18 KEGG enrichment pathways(P value<0.05),such Protein processing in endoplasmic reticulum and Glycolysis/Gluconeogenesis.At the same time,we also discussed and analyzed the genes which expressing top 10 under the temperature stress.(2)Based on the transcriptome,we cloned a heat shock protein 70 gene from MED by RT-PCR and RACE,we named it Btshp70.The complete cDNA of the genes was 2571bp,its opening reading frames was 1959bp and encoded 652 amino acids.Sequence analysis suggested that the Btsh70 shared high similarity with those HSP70 which were found in other cryptic species of B.tabaci.And it contained conserved motifys found in HSP70.There was no introns found in Bthsp70.(3)Among the differentially expressed genes,we found 11 fragments of HSP70.We used qRT-PCR to identify the relative expression level of mRNA of the 11 fragments which were under temperature sress.And found the results were consistented with results that measured by transcriptome.The expression levels of 11 HSP70 fragments which were found in transcriptome at 42? under high temperature stress were significantly different from the control.The result showed that HSP70 played an important role in the stress tolerance of B.tabaci,especially under high temperature stress.
