Potato virus Y(PVY)is a serious potato virus.All PVY strains could exist for a long time by infect potato tubers,which caused a variety of symptoms,resulting in a significant reduction in yield and quality.However,it is know that the mechanism of virus-host interaction in the process of potato tuber necrosis induced by PVY is still not clear.In this first study,we select PVY-infected potato tuber samples and healthy potato tuber samples,Four target genes: RCCR gene,LSD1 gene,St CaM2 gene and StCaM3 gene were obtained by gene sequence similarity comparison.First,the primers were designed and screened to the highly stable and specific primers.Then,four target genes were detected by real-time fluorescence quantitative PCR.The expressions of RCCR gene and LSD1 gene decreased obviously in the process of necrosis in potato tuber.The results showed that the LSD1 gene may be related to the necrosis of potatoes tuber,and the mechanism may be due to the decreased ability of LSD1 gene on CuZnSOD and CAT3.The structure of these two genes were analyzed,and the physical properties of RCCR gene and LSD1 gene were preliminarily determined.In the second study,the virus-free amaranth plant and the common tobacco plant were cultivated and the potato virus Y was purified to obtain a single PVY virus.Then in vivo PVY treatment,passivation test and protection test was carried out on 6 kinds of botanical insecticides tested by half leaf blight spot method,and using the system of host control effect in the host system test.The results showed that HN1 and HN5 were efficient in the protection test of Amaranth,and virus inhibition rate of HN1 and HN5 were 50.36% and 52.57%.In the passivation test of virus,the virus inhibition rates of HN4 and HN5 were 56.96% and 55.77% respectively.The inhibition rate of HN5 to potato Y virus reached 77.82% in the protection experiment,while in the PVY system host,although the control effect of HN5 was lower in the first survey,Thecontrol effect reached 63.41% in second survey.
