| Ethylalcohol extracts of fresh parsley roots、rhizoshperes and seeds have been examined after a repeated series of purification(from 1 to 4)in column chruomatography using a column with silicone coating.we screened the best fractions acording to count its allelopathic inhibition effect.To identify inhibitory allelochemicals released into the soil and seeds by parsley crop,ethylalcohol extracts of parsley fresh roots、rhizoshperes and seeds have been proceed with fifth and sixth chromatography(10 mm x 300 mm)fractions respectively by a column with silicone coating.It was used a bioassay that incorporated the different degree column fractions into PDA medium and co-cultured the plates with F.oxysporum f.sp.cucumerinum.The best fractions of ethylalcohol extracts of parsley fresh root were screened according to the allelopathic inhibition effect.Then further allelochemicals of 12 best fractions of the sixth chromatography were identified by GC-MS,so as to determine the compositions,name and molecular structure of the allelochemicals in the best fractions.Finally the induced resistanc of best fractions with the method of root-irrigation on the F.oxysporum f.sp.Cucumberinum were studied by using field experiment.The research results were as following:1.12 best fractions were obtained form 3 kinds of parsley materials(parsley fresh roots、rhizoshperes and seeds)extract solution after a repeated series of purification(fifth and sixth cycles)in chromatography.There were significant diferen allelopathic effect for the degree column fractions obtained from the fith and sixth cycles of chromatography respectively on F.oxysporum f.sp.Cucumerinum.The sixth optimal fractions of ethylalcohol extracts of parsley fresh root is RE2110186、RE295548、RE623597 and RE686568,the allelopathic effectl compared with the sixth cycle tomographic distilled water(the control,RCK5-8)was 78-14%、78.83%、73.37%and 71.34%,respectively,and the allelopathic effect 2 compared with the sixth cycle tomographic ethylalcohol(the control,RECK5-8)was 70.14%、71.38%、64.84%and 60.64%,respectively.The sixth optimal fractions of ethylalcohol extracts of parsley rhizoshpere is RSE222296、RSE233667、RSE459638 and RSE499152,the allelopathic effectl compared with the sixth cycle tomographic distilled water(the control,RCK5-8)was 73.90%、77.59%、77.05%and 73.65%,respectively,and the allelopathic effect 2 compared with the sixth cycle tomographic ethylalcohol(the control,RSECK5-8)was 64.50%、68.70%、68.34%and 64.24%,respectively.The allelopathic effect1 and allelopathic effect2 of the sixth optimal fractions of ethylalcohol extracts of parsley seed(SE138648、SE189357、SE819257 and SE8411085)compared with the sixth cycle tomographic distilled and ethylalcohol water(the control,SCK5-8 and SECK5-8)was 70.53%、74.58%、69.70%and 70.11%as well as 61.71%、65.31%、61.88%and 59.27%,respectively.2.Allelochemicals were found in 12 best fractions of ethylalcohol extracts of parsley fresh root,rhizoshpere and seed using GC-MSThere were 11 kinds of allelochemicals from the best fractions of ethylalcohol extracts of parsley fresh root contained in phenol,ester,amine and alkanes 4 classes in the GC-MS analysis.They were 2,4-bis(1,1-dimethylethyl)-Phenol,,8,11-Octadecadienoic acid,methyl ester,Ethyl 13-docosenoate(ethyl erucate),Dimethyl phthalate,1,2-Benzenedicarboxylic acid,butyl 2-methylpropyl ester,1,2-Benzenedicarboxylic acid,butyl 2-ethylhexyl ester,Phthalic acid,butyl 2-pentyl ester,Dibutyl phthalate,9-Octadecenamide,(Z)-,Heptacosane,3-ethyl-5-(2-ethylbutyl)-.There were 6 kinds of allelochemicals from the best fractions of ethylalcohol extracts of parsley rhizoshpere contained in phenol,ester,acids and amine 4 classes in the GC-MS analysis.They were 2,5-bis(1,1-dimethylethyl)-Phenol,1,2-Benzenedicarboxylic acid,butyl octyl ester,2.3-Dihydroxypropyl cis-13-docosenoate,Phthalic acid,butyl isohexyl ester,Erucic acid and 9-Octadecenamide,(Z)-.There were 7 kinds of allelochemicals from the best fractions of ethylalcohol extracts of parsley seed contained in ester and alkanes 2 classes in the GC-MS analysis.They were(E)-9-Octadecenoic acid ethyl ester,1,2-Benzenedicarboxylic acid,diisooctyl ester,13-Docosenoic acid,methyl ester,(Z)-,Phthalic acid,6-ethyl-3-octyl butyl ester,Octadecane,Eicosane and Heptadecane,9-hexyl-.Based on the compositions of allelochemicals in each best fractions,esters should be the main allelochemicals in the parley extract solution.3.The induced resistanc of the best fractions of ethylalcohol extracts(RE2110186、RE295548、RSE233667、RSE459638)of parsley fresh root、rhizoshpere with root-irrigation for Cucumber seedlings were studied,the results showed that cumber seedlings resist to the cucumber wilt was induced by all of 4 best fractions.The best fractions(RE295548 and RSE459738)of ethanol of parsley fresh root and rhizoshpere have the highest induced resistance effects in the 4 best fractions.The induced resistance effectl and induced resistance effect2 of the best factions of RE295548 from ethanol of parsley fresh root reached to 61.53%and 55.33%,respectively,and the induced resistance effectl and induced resistance effect2 of the best factions of RSE459738 from ethanol of parsley rhizoshpere reached to 58.17%and 51.43%,respectively. |
