| Agropyron mongolicum belongs to gramineae perennial grasses of wheat,which contain abundant protein,and it is well of palatability.It is an excellent grass and it has very high ecological value and the forage value.Agropyron mongolicum seed production reduction caused by the phenomenon of shattering is very serious.In order to reveal the mechanism of shattering in Agropyron mongolicum,we cloned the genes controlling the rate of shattering in Agropyron mongolicum by homologous cloning,and detected the expression of these genes in different tissues of Agropyron mongolicum.The main conclusions are as follows:1.The Amsh1-1 gene 935bp of Agropyron mongolicum cDNA.5 ’non coding region(UTR)127bp,the’ 3 ’non coding region(UTR)247bp,open reading frame(ORF)561 bp,encoding a total 186 amino acids.2.The Amsh1-2 gene 1118bp of Agropyron mongolicum cDNA.5 ’non coding region(UTR)247bp,the’ 3 ’non coding region(UTR)247bp,open reading frame(ORF)624bp,encoding a total 207 amino acids.3.The Amsh1-1 gene of Agropyron mongolicum was the highest homology with the Hvshl gene of barley,while Amsh1-2 gene had the highest homology with bamboo.4.The expression of Agropyron mongolicum roots,stem and leaf,panicle of different tissues such as the most stable reference genes for ACT and APRT.the most stable reference genes for TUBβ5.Amsh1-1 and Amsh1-2 were mainly expressed in the spike of Agropyron mongolicum,followed by stem,and the lowest expression was leaf and root. |
