Japanese encephalitis(JE)is a zoonosis caused by Japanese encephalitis virus(JEV).As a kind of neutropic virus,JEV,spreading by mosquito,can invade the central nervous system(CNS)through the Blood-brain barrier(BBB),leading to severe neuroinflammation and death.JEV peripheral immune escape is the leading cause of its invading the CNS,but the mechanism of its escaping from the peripheral immune system is not clear.In this study,the model of C57BL/6 mice infected with JEV-P3 was used to explore the function of MDSCs during the process of JEV peripheral immune escape.C57BL/6 mice were infected with 105 PFU of JEV intravenously(i.v.).On 1-5 days post infection(d.p.i),serum and spleen tissue were collected every other day for detection of MDSCs related cytokines.It was found that the number of MDSCs increased in spleen on day 3 and its immunosuppressive molecules rised accordingly at the same time.Arg-1 was detected by ELISA,we found that Arg-1 which was expressed by the MDSCs showed a obvious rising trend under the JEV stimulation.We applied relative fluorescence quantitative polymerase chain reaction(PCR)to detect the cytokines during the differentiation of T cell and found that the T cell differentiation was inhibited while the MDSCs increased.ADAM17 protein and CD62 L molecules were detected by flow cytometry,the results showed that the expression of ADAM17 went up while CD62 L expression decreased.In summary,our study demonstrates that JEV can escape the peripheral immune response and invade into CNS via the suppression function of MDSCs.
