Plant trichomesare special epidermal cells,which extends outward to form a hair like structure.Plant trichomesareevoluted to conform biological and abiotic stress.They are widely distributed on the surface of the terrestrial plants.Plant trichome is seem as the basis of plant somatic cell differentiation model,there are more and more researches about theregulatory mechanism among the different plant trichome,new gene discovery and function analyze are expanding.At present,the research about related genes of Arabidopsis' s single cell surface isthe most thorough.However,the development mechanism of tomato,tobacco and other multicellular trichome is very different from Arabidopsis,because of their different cell structure.Sl HZ45 gene of tomato can promote the development of type I and type ? trichomes significantly,and promote the development oftype ? trichomes highly significant.Therefore,the separation and functional study of Sl HZ45 gene will play an important role in the development and regulation mechanisms in multicellular trichomes.Because of the special morphological structure and the secretion of secondary metabolites,Plant trichomes play a role in the stress resistance.The types of trichomes which is promoted by Sl HZ45 gene are all gland trichomes.Therefore,it is determined thatfuction of Sl HZ45 gene is about the resistance to adversity.And this research can provide good material for resistant breeding of tomato.Based on the established tobacco Sl HZ45 gene transformation system,the tomato plant regeneration transformation,epidermal hair change analysis and expression of the gene analysis of experimental physiology,stress related gene,determine the function of Sl HZ45 gene.The main results are as follows:1.The transformation system of tomato Sl HZ45 gene was constructed,and the types and numbers of trichomes were compared with that of the common tomato cultivars.The results showed that compared with the control group,the number of type?and type ? trichomes on the leaves of Sl HZ45 transgenic tomato plants was increased by 45.80% and 50%,respectively,and all of the twotrichomes were glandular hairs.Compared with the control group,the number of trichomes on the stem surface of Sl HZ45 transgenic tomato plants was significantly increased,the growth rate was 395.84%,and the type of the same as the glandular hair.Flower trichomes compared with control group,both the number and type of petals,receptacle trichomes were no significant difference.The number of trichomes on the surface of the Sl HZ45 gene showed an overall increasing trend,and the total number of epidermal hairs increased by 48.66%.2.T1 generation Sl HZ45 transgenic tobacco trichome types analysis of Sl HZ45 gene has significant effect to promote the occurrence of tobacco trichomes,while the overall number of promoting effect is not obvious,and the over expression of tomato trichomes occurred on the type of agreement,Sl HZ45 gene can significantly promote plant trichomes the occurrence of occurrence and inhibition of some non-glandular hairs.3.The design of full-length primers on tomato plants to cold,salt and drought stress in different parts of the time gradient sampling,semi RT-PCT and Real-time PCR analysis results showed that the expression of Sl HZ45 gene in different parts of both in root and fruit was very low,the highest expression level in flower.3 kinds of gene expression of stress in root and fruit is still relatively low,and the peak value of 6h was the highest in the gene expression in flower organ of salt treatment,cold stress middle leaf and flower were the peak at 3h,and the relative expression close to the expression peak of 6h in under drought stress.The expression of SlHZ45 gene in stem,leaf and flower increased significantly with the prolonging of stress treatment time in 3h-6h.At the same time,transgenic T1 generation tobacco was treated with three kinds of adversity stress,which was consistent with the expression of the gene in tomato.4.150 mM salt and 20%PEG processing of T1 generation tobacco seed germination experiment,the results found in the seed germination experiment,compared with the control group,SlHZ45 transgenic tobacco seed germination rate with the stress increased,the efficiency and the final germination rate were significantly higher than the control group.It can be inferred that Sl HZ45 gene plays a certain role in the resistance to adversity in seed germination stage.5.In the determination of root length,the root length of transgenic tobacco was higher than that of the control group 0.5cm,and the lateral root branch was more than the control group.Under salt stress,transgenic tobacco roots were longer than those in control group,and the lateral root branches were higher than those in control group.Under drought stress,the root length was similar with salt stress,the root of transgenic tobacco was higher than that of control group,and the lateral root was more than that of control group.6.The promoter sequence of Sl HZ45 gene prediction and bioinformatics analysis,using the online software results showed that the promoter sequences except core promoter elements and CAAT box,and many reported cis acting regulatory elements.Such as: AE-box,GA-motif,GAG-motif,TCT-motif,ATGCAAAT,motif,Skn-1-motif,Circadian,TC-rich,repeats-motif,HSE-motif,GARE-motif,GAGTA-motif etc.It is suggested that the Sl HZ45 gene is likely to be expressed by multiple transcription signals.
