Spermatozoa cryopreservation was the best technology to store boar sperm for a long-term period,which is a vital way to save valuable genetic material,This technique provided the accessibility to spermatozoa,despite of time and location.Compared with fresh spermatozoa,frozen-thawed spermatozoa had a lower motility and fertilizing capacity,which largely restricted the extensive application of cryopreserved boar sperm.In present study,semen samples were collected from mature Duroc boars by the gloved hand technique and frozen–thawed in the extender with various concentrations of lycopene?sesamol and baicalein.Semen samples were assessed for spermatozoa motility,plasma membrane integrity [hypoosmotic swelling test(HOST)],mitochondrial activity,acrosome integrity,DNA integrity,lipid peroxidation and Superoxide dismutase(SOD),Catalase(CAT),Glutathione peroxidase(GSH-Px)activities after thawing.The results showed that:1.Motility,mitochondrial activity,plasma membrane integrity,acrosomal integrity and DNA integrity of frozen-thawed boar spermatozoa were significantly improved by added different concentrations of lycopene in freezing media,compared to those of control(P<0.05).Higher motility,mitochondrial activity,plasma membrane integrity,acrosomal integrity were achieved when lycopene was added at 2.0?mol/L(P<0.05),and a higher DNA integrity was 78.69%,when the concentration of lycopene was 3.0?mol/L(P<0.05).2.The sperm quality parameters of frozen-thawed boar spermatozoa were observably improved by supplemented with appropriate concentrations of sesamol in boar freezing media(P<0.05).The optimum concentration was 2.0g/L(P<0.05).No significant difference was observed between 0.05g/L group and the control(P>0.05).3.The obviously protective effect was obtained with the addion of various concentrations of baicalein in boar freezing extender(P<0.05).Better effects were presented when the concentration of baicalein was 0.04g/L and 0.06g/L(P<0.05).4.The better cryopreservation result was gained with the addion of lycopene and sesamol,due to they had a synergistic effect.The superior combined consistency was 0.2g/L sesamol and 2.0?mol/L lycopene(P<0.05).5.The MDA level of frozen-thawed boar spermatozoa was significantly reduced,and the enzyme activities(SOD ? CAT and GSH-Px)were apparently increased with certainconcentration lycopene,sesamol and baicalein added in the boar freezing media(P<0.05).The better concentrations of lycopene,sesamol and baicalein were 2.0?mol/L ? 0.2g/L and 0.04g/L(P<0.05),respectively.The antioxidant ability of frozen-thawed boar spermatozoa was further increased by combined use 0.2g/L sesamol and 2.0?mol/L lycopene,0.2g/L sesamol and 2.5?mol/L lycopene(P<0.05).Conclusion: Three kinds of antioxidants had obvious protective effect to boar spermatozoa in the freezing and thawing process.A better effect was obtained by combined utilization of lycopene and sesamol.
