Moso bamboo is the high yield of forest resources in our country,with large grow area and widely used,it occupies a very important position in the bamboo industry.Since it has high economic,ecological value and closely connected with people's life,so it is loved by people.So the research of moso bamboo has very important practical application value.Previous studies of moso bamboo,mainly focus on morphology,ecology and nutrition,but about the internal molecular mechanism research of growth and development remain unknown.About before and after the beginning of autumn winter shoot develop from bamboo rhizome lateral bud and rhizome shoot as the apex of the bamboo rhizome organization.The two tissues in moso bamboo in the process of rapid growth and bamboo rhizome rapid extension have important biological significance.So it is the first time we build about moso bamboo shoot tip and rhizome shoot tip tissue's chromatin immune coprecipitation sequencing library(ChIP-seq),and analysis of two kinds of the most common with the most important two types of histone modification:H3K4me3 and H3K27me3.we optimize the process of library building and establish a suitable ChIP experiment method for moso bamboo.Through bioinformatics analysis,reveals the shoot tip and rhizome shoot tip's modification of histone H3K4me3 and H3K27me3 genome-wide distribution characteristics and differences.After Peak-calling to the sequencing results,we get a total 5,379 sequencing Peaks,after comparing the peak to bamboo gene annotation database,and wipe off the same gene with different modification site situations,finally we get 3,293 independent modified genes.The rhizome shoot tip peculiar has 219 H3K4me3 modified genes and 827 H3K27me3 modified genes.Bamboo shoot tip peculiar has 1,105 H3K4me3 modified genes and 569 H3K27me3 modified genes.There are 539 two kinds of modified genes.32 genes have 3 kinds of modification,4 kinds of modified genes have 2.This result suggests that histone modification sites of rhizome shoot tip and bamboo shoot tip in the genomes of bamboo distribution has tissue specificity,universality and diversity.From the perspective of peaks combine with upstream and downstreamgenes,thebamboo shoot tip and rhizome shoot tip's H3K4me3 modification peak signal mainly distribute near the transcription start site and transcription termination site and few within genes,at transcription start site bamboo shoot tip signal to be stronger than rhizome shoot tip's,but near the transcription termination site bamboo shoot tip signal to be weaker than rhizome shoot tip's.H3K27me3 modified peak signal is also appear near the transcription start site and transcription termination site,rhizome shoot tip's signal have always been stronger than bamboo shoot tip's.Through use the Cytoscape software to do GO analysis of bamboo shoot tip and rhizome shoot tip's H3K27me3 modified genes,we build the genes function enrichment graph.Through the study of its function annotation,we find that most H3K27me3 modified genes locate in the nucleus and mainly involve in the regulation of the transcription of DNA regulation area binding protein,transcript factors transcription process and its activity,DNA and RNA mediated expression of other genes transcription process.Suggest that histone modification can be realized through regulating the above-mentioned genes to regulate the expression of other genes.From the preliminary work we do,can lay a solid foundation to subsequent research of bamboo epigenetic regulation.
