Trematode is a kind of parasite which was mainly parasitic in the host bile duct,intestinum tenue and other parts,and is endemic wordwide.It can cause huge economic losses.Zoonotic parasites cause serious threaten to human health,and even threaten life.Ribosomes widely exists in cells,plays an important role in the life activities,it is also an important place for protein synthesis,ribosome DNA is the gene which encording ribosomal RNA.It consisting of transcribed ribosomal RNA and non transcribed spacer.Through the ribosomal sequence,the species can be identified,the taxonomic status of species,the diversity of species can be determined,etc… Therefore,this study amplified the complete ribosomal sequences of male and female Schistosoma turkestanicum,Metorchis orientalis,Clonorchis sinensis,Echinoparyphium recurvatum,and Echinostoma hortense by the methods of RCR,which cause serious harm to human and livestock.And phylogenetic tree based on different marker genes.The complete ribosomal sequences of five trematodes were 8 209~8 917 bp in length,and the 6 regions of ribosomal sequences almost no variation in length,there was only a slight change in length of IGS.By comparing the homology of 5 species,the homology were 89.7~99.8% for 18 S,56.4~94.8% for ITS1,92.4~100% for 5.8S,49.5~96.2% for ITS2,80.3~96.8% for 28 S,34.2~94.1% for IGS.It can be seen that the intraspecific variations was low,and the interspecific variations was high.At the same time,according to the interspecific variation,we can get the six gene conservative sort,and they should be: 5.8S>18S>28S> ITS1>ITS2 >IGS.Phylogenetic tree based on different marker genes,the result is not entirely consistent with the traditional taxonomy.Based on the phylogenetic tree constructed by 18 S and 28 S,the evolutionary relationship in families and orders is inconsistent with traditional taxonomy.Based on the phylogenetic tree constructed by ITS1 and IGS,the evolutionary relationship in generas is inconsistent with traditional taxonomy.The phylogenetic tree based on ITS2 is consistent with the traditional taxonomy.It shows that 18 S,ITS1,ITS2,28 S,IGS are suitable for phylogenetic studies among orders,families,genera,orders,families,respectively.We can infer that IGS can be used as a gene marker to explore the evolutionary relationship of the flukes.The complete ribosomal DNA sequences of 30 samples from 5 species were firstly obtained.These findings enriched information of trematode ribosomal genome dataset.We get the conservative sort of each ribosomal gene.The phylogenetic trees constructed by using different ribosomal genes showed that different marker genes were suitable for different taxonomic classes. |
