Peste des petits ruminants(PPR)is an acute and highly infectious diseases of small ruminants including sheep and goats,which is caused by Peste des petits ruminants virus(PPRV).The outbreak of the PPR among sheep cccurred in a farm in the south Xinjiang of Aksu region during May 2014.Among those sheep mang of them appeared high fever,Depressed of spirit,gray stench secretions in the Mouth and nose,dyspnea,diarrhea,angular and so on.Through the clinical diagnosis and pathology examination showed that the sheep had PPRV infection Initially suspected,By Formaldehyde Solution fixed lung tissue nucleic acid extraction and PPRV N gene sequence amplification,The results of PCR sequencing were compared with Blast,The homology of the gene fragment with the second PPR epidemic strains in China was as high as 99%,We Confirmed that the sheep farm was infected with PPRV.The laboratory extracted Nucleic acid from Formaldehyde Solution fixed lung tissue and amplified PPRV gene sequence.The whole gene sequence of Xinjiang PPRV strain was obtained and named as China XJNJ2014,According to sequence analysis of N F gene and the Whole gene sequence,the results showed that The homology of test strain had the highest with GZL-14 strain in Jilin,and the highest homology with Izatnagar / 94 strain and Sungri 1996 MSD strain in India,Nucleotide sequence of N gene analysis showed The relationship of test strain between the China/XJYL/2013 strain and the Guangdong GD/QY/2014 strain had Evolutionary relationships,With foreign India/Delhi/2016/05 strain had Evolutionary relationships.Nucleotide sequence of F gene analysis showed The relationship of test strain between the China/BJ/2014PPRV-FY and GZL-14 strain had Evolutionary relationships,With foreign Izatnagar/94 and Izatnagar-94 strain had Evolutionary relationships.Genome and N,F gene nucleotide analysis showed that the test strain was far from the evolution of the Tibetan strain and the vaccine strains currently used in China.Protein analysis of N showed The relationship of test strain between the CH/HNNY/2014 the CH/HNZM/2014GD/QY/2014 PPRV-FY and IND/Delhi/2016/05 strain had Evolutionary relationships.Protein analysis of F showed The relationship of test strain between the CH/HNZK/2014 the China/BJ/2014 GZL-14 PPRV-FY and Izatnagar/94 strain had Evolutionary relationships.N,F Protein suquence analysis showed that the test strain was far from the evolution of the Tibetan strain and the vaccine strains currently used in China.The whole genome and N,F gene nucleotide phylogenetic tree showed that the tested strain had a small branch with the second epidemic virus strain in China,and it was close to the evolution of the Indian strain.This study showed that the epidemic of this disease is caused by PPRV infection in sheep in Xinjiang Aksu region.The results of gene sequence analysis showed that the test strains were closely related to the evolution of the strains during the PPR pandemic in China 2013,Phylogenetic relationships revealed that the test strain was in the same branch as the pandemic strain(IV),This study can provide reference for the diagnosis of PPR in this region,it can provide a theoretical basis for the prevention and control of the epidemic.
