| Somatic embryogenesis have great theoretical significance and applied value.It’s not only used in mass propagation and germplasm conservation,but also attacted in quality breeding and transgenic receptor system optimized with genetic engineering technology.In this study,we used five different varieties of Lilium as materials and studied effects of different explants,different concentrations of PIC and culture conditions on embryogenic callus and somatic embryogenesis.That laid the foundation for establish efficient somatic embryogenesis and rapid propagation system and genetic transformation of Lilium.The main results are showed as follows:1.Leaves were used as explants to induce somatic embryogenesis:It was found that somatic embryos induced by leaves as explants were first to form embryogenic callus,and then somatic embryos were differentiated from embryogenic callus.The results showed that optimal medium for inducing embryogenic callus from ’Strawberry and cream’ leaves was MS+2.0 mg·L-1 PIC+0.2 mg·L-1 NAA and other four cultivars was MS+1.5 mg·L-1 PIC+0.2 mg·L-1 NAA and the effect was better under condition of dark culture.The induction rate of’White heaven’ and ’Triumphator’ could reach up to 95.83%,and induction rate of ’Eyeliner’reached 95%,and induction rate of ’Strawberry and cream’ and ’Double surprise’ were 93.33%.2.Scales were used as explants to induce somatic embryogenesis:Somatic embryogenesis formed different way by scales as explants.The results showed that optimal medium of ’Eyeliner’ and ’Double surprise’ were MS+2.5 mg·L-1 PIC+0.2 mg·L-1 NAA and culture condition was dark culture,the induction rate was 90.83%.The optimal medium of’White heaven’ was the same as ’Eyeliner’,but effect was better under condition of light culture and induction rate was 96.67%.The optimal medium of ’Strawberry and cream’ and’Triumphator’ was MS+2.0 mg·L-1 PIC+0.2 mg·L-1 NAA."Strawberry and cream" had better induction effect under light culture condition and induction rate was 88.33%.’Triumphator’had better induction effect under dark culture condition and induction rate was 93.33%.3.The embryogenic callus was transferred to the differentiation medium.The results showed that the dark culture was more favorable to the differentiation of embryogenic callus and the formation of somatic embryos.The rate of ’Triumphator’ differentiation was 97.5%.The rate of ’Eyeliner’ and ’Strawberry and cream’ differentiation were 95%.The differentiation rate of ’Double surprise’ and ’White heaven,respectively were 92.5%and 90%.4.The embryogenic callus with different morphological somatic embryos were transferred to medium MS+0.5 mg·L-1 BA for germination and we found that light culture is more advantageous.The germination rate of ’Eyeliner’and ’Triumphator’ could be reached 97.5%.The germination rate of ’White heaven’ and ’Double surprise’ could be reached 97.5%.The germination rate of ’Strawberry and cream’ reached 92.5%.5.The regenerated plantlets grew vigorously with 1 perlite:1 peat:1 vermiculite.The survival rate of ’Eyeliner’ and ’Triumphator’ can reach 97.5%.’White heaven’ and ’Double surprise’ survival rate achieve 95%and ’Strawberry and cream’ survival rate is relatively low,up to 92.5%. |
