Zn2Cys6 transcription factor family belongs to C6 transcription factor in zinc cluster family,which is a common type of fungal transcription factor.The first study of this family transcription factor is the GAL4 gene in Saccharomyces cerevisiae,and there are more studies in plant pathogenic fungi,such as Magnaporthe oryzae and Fusarium graminearum.The study found that the transcription factor is involved in the life processes,such as sporulation and pathogen growth.Alternaria brassicicola caused the black spot of cruciferous plants,bringing serious economic losses to farmers.At present,the pathogenesis of A.brassicicola is not clear.It is found that the transcription factor of Zn2Cys6 family is involved in the pathogenesis of many Phytopathogenic fungus.There is a little studies on A.brassicicola.Therefore,in order to help us understand the pathogenesis of the bacteria,we have to study deeply.In this paper,we selected the MoNIT4 gene of the Zn2Cys6 family from Magnaporthe grisea and found its homologous gene in A.brassicicola,.Based on the principle of homologous recombination,the mutant of ZnCysAb0012 gene was obtained by protoplast transformation method.And the function of ZnCysAb0012 gene in A.brassicicola was studied.Through the comparative study of ZnCysAb0012 deletion mutants and wild type,we found that the growth rate of ZnCysAb0012 deletion mutants was 1.96 times faster than that of wild type.Through the observation of two kinds of fungus,it was found that the main growth of wild type is buried hypha,and the main growth of ZnCysAb0012 gene deletion mutant was aerial mycelium.And difference of the colony color is very large.ZnCysAb0012gene deletion mutant is gray,while the wild type is dark green.Through the inoculation experiments on rape,it was found that rapeseed inoculated with the ZnCysAb0012 gene deletion mutant was not pathogenic to leaves.Through the microscopic observation of the cultured strains,we found that the ZnCysAb0012 gene deletion mutant strain did not have spores under the microscope.At the same time,through the study of penetrating ability andresistance to stress,ZnCysAb0012 gene deletion mutant was not significantly different from wild bacteria.In this summary,ZnCysAb0012 gene can control the spores of pathogens,and also has a significant effect on the growth rate and pathogenicity of pathogens.While the ability to penetrate the pathogen and the ability of adversity to grow is relatively small.
