Selenium is a very important trace element in human and animal body,for the body's health has a variety of biological functions.Selenium in nature there are two forms: inorganic selenium and organic selenium,and organic selenium compared to inorganic selenium a wider range of safety,but also more nutritional value.By applying the inorganic selenium to crops,crops to transform inorganic selenium into organic selenium,and then enter the food chain,make human body to achieve the purpose of filling selenium.Peanut is one of the most important oil crops and cash crops in our country,and it is not only of high nutritional and health benefits,but also of high selenium enrichment.In this paper,black peanut was used as the research object,and the physiological characteristics and selenium content of black peanut were studied.And the different forms of selenium in peanut were analyzed and the following conclusions were drawn:The effects of three kinds of selenium application methods and different Se gradient on the physiological characteristics of peanut were also investigated.From the point of view of selenium application,leaf selenium is the best,the soil foliar is the second,and the soil is the worst.From the selenium gradient,for most of the growth indicators of peanut(budding rate,plant height,yield)with the trend of growth and then decrease.Excessive selenium concentration will produce toxic effects on peanuts,resulting in peanuts burning or even death and so on.Selenium concentration had no significant effect on the growth index of peanut plumpness and chlorophyll content.It was determined that the best application of selenium was the application of selenium in the leaves,and the best concentration of selenium was about 1.2mg/L.When the concentration of more than 4mg / L,will produce the peanut toxic,affecting the normal growth of peanuts.The selenium in peanut and peanut leaves of selenium-enriched peanut samples were determined by microwave digestion-hydride generation-atomic fluorescence spectrometry.The effects of different oxidants and microwave digestion conditions on the digestion of selenium-enriched peanut were studied.The optimum oxidant system was determined as follows: HNO3-H2O2(5mL: 2mL);The best Microwave Digestion procedure:(1)1 min? 0.3Mpa;(2)3 min? 0.6 Mpa;(3)3 min?0.9 Mpa;(4)2 min?1.3 Mpa;Black peanut has a strong ability to enrich the exogenous selenium,and selenium application on the leaves is more conducive to the enrichment of selenium in black peanut,with the increase of selenium concentration,the selenium content in peanut also increased,but the selenium-rich multiples are lower,shows that the low concentration of selenium can promote toabsorb seleniumfor peanuts,with the increase of Se enriched nutrient solution,the absorption of Se in peanut was inhibited.The distribution of selenium in peanuts is also very different,the selenium content of peanut leaf is far greater than the amount of selenium in peanut.0.1mol/L NaOH;0.1mol/L HCl;Ph 7.2Tris-HCl;Deionized waterwere used as extraction solution to extract the soluble Se and Hydride Genernation(HG)-AFS with Microwave was used to determine the Se content in the peanut.The results show that the order of selenium content in four type of extraction was:NaOH >H2O >HCl> Tris-HCl.The result show that mostly of selenium was dissolved in NaOH,which account for about 32 % in the total selenium.The proportion of selenium in the water and HCl extraction respectively were 20 % and 15 %.The result show that the organic selenium is the main form in the peanut.Alkali soluble acid isoelectric point method was used to extract the soluble Se in the Se-enriched peanut,the result showed that the optimal condition forextraction was:isoelectricpoint,pH=4.1.extract concentration,0.23mol/L;material/liquid ratio,1:23;extraction time,11.2h;extraction temperature,53?.The results showed that the Se content in the protein increased across the concentrations of Se in the solution,therefore,a high concentration of Se can not only increase the Se content in protein,but also the proportion of total Se content in protein.The objective of this study was to develop a high performance liquid chromatography-tandem mass spectrometry(HPLC-MS)method for the determination of selenium-rich peanut selenomethionine(SeMet)content approach.The samples of selenium-rich black peanut were extracted by Tris-HCl solution at PH 7.2 with acetonitrile: water ratio of 70:30 as mobile phase,and the filtrate was isolated by Agilent Bond Elut Plexa column by online electrospray ionization source ESI positive ionization after using multiple reaction monitoring mode MRM select m/z198.0 ? 109.0,m/z198.0 ? 181.0and m/z198.0 ? 56.0 determination SeMet of selenium-enriched black peanut.The liner range of selenomethionine was between 0-250 mg/L,the accuracy was between 96.1%~101.1%,the RSD value was within 5%,SeMet detection limit was 0.05769ng/g.The results showed that: The method is sensitive,high precision,specificity,and the treatment of sample is simple.So,it can be used to determine the content of SeMet in selenium-enriched black peanut.
