Grass carp (Ctenopharyngodon idella), a most important aquaculture fish in China, is susceptible to hemorrhage disease. The disease is caused high mortality to one year old grass carp. The mature grass carp, survived after infected by GCRV at one year old, was used of blood fish. The eggs, fertilized egg, embryo, larvae, and serum of blood fish of three groups, Treated ((?))×Treated ((?))(group P), Treated ((?))×Control ((?))(group M), and Control ((?))×Control ((?))(group C), was collected at different time points. The measure of enzyme-linked immunesorbent assay (ELISA) and turbidimetric inhibition immune assay was used to detected the activety of IgM, C3, LZM in the serum, egg, fertilized egg, embryo and larvae. Meanwhile, the relative expression of IgM, IL-1?, IFN, TLR3, and TLR22 mRNA in spleen, trunk kidney, head kidney, and thymus of six-month old grass carp were detected by real time fluorescence quantitative PCR (RT-qPCR) after infected with GCRV at different time point. Results as follows:1 IgM, C3, and LZM activity in serum, eggs and embryos:In this study, the IgM activity of blood fish serum and fertilized egg were 2.7-fold and 1.3-fold to the control group, respectively. In the embryo of immune blood fish, the IgM activity higher and longer protect time than control group. Compared with group M, the IgM activity higher in group C. The C3 activity of group C and group M have no signification with control. In the early stage of onto, group P has signification advantages in lysozyme activity, but the activity no signification different after hatched.2 Relative expression of maternal immune related gene infected by GCRV in offspring of grass carp:Intraperitoneal injection GCRV 200?L per tail, the IgM mRNA relative expression in the spleen, trunk kidney and head kidney of group P and group C higher than group C. In thymus of three groups, the relative expression of IgM mRNA were no significant different. The interferons mRNA relative expression in trunk kidney and head kidney higher than group C, but the relative expression of group P no significant difference with the group C. The IL-1 beta mRNA relative expression of group M in head kidney, spleen and trunk kidney higher than the group C, but the expression of group P no obvious advantage than the group C. In the immune response of grass carp resisted GCRV of three kinds of grass carp, TLR22 mRNA relative expression was no significant difference.Conclusion:The LZM and IgM activity in mature eggs or embryos of grass carp can be improved by increased the LZM and IgM activity of blood fish. Meanwhile, the maternal immune related gene (IgM, interferons, IL-1 beta and TLR3) mRNA expression level can be improved by immune blood fish.
