The Expression GmLEC1 And GmFUS3 During Seed Development And It's Relation Ship With Storage Protein Synthesis In Soybean

Soybean as an industrial crop in the world, is a major source of protein in humans and animals, contains storage protein nutritional value is high, the application prospect is very broad, but the regulation mechanism of soybean seed storage protein synthesis is unclear. The paper tested the protein content of soybeanSCS9299RR seed in different development periods through kjeldahl method, based on the regulatory networks that control seed storage proteins synthesis in Arabidopsis, and studied the expression of GmLEC1-a,GmLEC1-b,GmFUS3-16 and GmFUS3-19 by RT-PCR and RT-qPCR. To exploring the regulation mechanism that control seed storage protein synthesis in soybean then we analysed the correlation of the gene expression value with Gy1,CG-2. The main results from the study as fowllow:(1) By kjeldahl method we tested the protein content of soybeanSCS9299RR seed in different development periods and obtained protein variation trend during seed development and maturation. And then we combined microscopically the expression results of Gyl, CG-2,showed that soybean seed storage proteins synthesis in about 30-50 days after flowering.(2) The expression level of LEC1 in different tissues of soybeanSCS9299RR by RT-PCR showed GmLECl-a,GmLECl-b only specifically expressed in soybean seeds. The result of RT-qPCR showed the expression patterns of GmLEC1 -a was consisten with GmLECl-b, peaked at 30DAF.(3) The expression research result of GmFUS3-16 and GmFUS3-19 in soybeanSCS9299RR showed GmFUS3-19 had different expression in developmental periods, peaked at 40DAF,and its pattern has some similarities with LEC1.But GmFUS3-16 may not be expressed in seed development and maturation,may not involved in the seed storage protein synthesis of soybean.(4) By bio-statistical methods for analysis of gene expression value, the result showed:GmLECl-a with Gy1,CG-2 were moderate non-significant positive correlation; GmLECl-b with Gyl,CG-2 were moderate non-significant positive coTrelation;GmLECl-a,GmLECl-b with GmFUS3-19 were moderate significant positive correlation; GmFUS3-19 with Gyl,CG-2 were highly significant positive correlation, described LEC1 may depend on FUS3 regulate and control seed storage protein synthesis in soybean.In brief, soybean seed storage protein synthesis and accumulation is a process that was regulated fine and strictly,and have a certain law. As transcription factor genes, GmLEC1-a,GmLEC1-b and GmFUS3-19 have some correlation with the expression of storage protein genes, speculating that they might be closely related to seed storage proteins synthesis in soybean.
