Lycopersicon esculentum Mill is an annual herb, belongs to solanaceae, has the characteristics of rich nutrition and unique flavor. Apium graveolens belongs to umberlliferae, rich smell, can be used for seasoning. Vigna unguiculata also is an annual herb, belongs to leguminous, play an important role in exportation agricultural products. These three kinds of vegetables are rich in therapeutic value, and they are popular with people. In recent years, with the increase of market demand, these three vegetables'planting area are growing, the popularity of plant virus disease is increasing. In August 2014 in Shanxi agricultural university, the Lycopersicon esculentum Mill with mosaic symptom, Apium graveolens with dwarf symptom, Vigna unguiculata with yellowing and mosaic symptoms were found. In order to identify the viral pathogens that induced the disease of these three vegetables, biological inoculation, extraction of double-stranded RNA (dsRNA), sequence-independent amplification (SIA) and RT-PCR method were used.(1) Identification the pathogens that induced their disease by biological inoculation, the results showed that their disease was likely to be caused by viruses.(2) dsRNA were extracted from leaves of infected plant samples and used as temples in SIA, results of PCR and sequencing showed that the viral pathogen infect Lycopersicon esculentum Mill was Cucumber mosaic virus, named XHS_CP, the viral pathogen infect Apium graveolens was Bell pepper endornavirus, named BPEV-QC, and the viral pathogen infect Vigna unguiculata were Phaseolus vulgaris endornavirus 1 and Bean yellow mosaic virus, named PvEV-JD and BYMV-JD, respectively.(3) Analysis those viruses' sequences and phylogenetic trees showed that XHS_CP isolate belongs to CMV subgroup I, the amino acid sequence of CMV CP gene were conservative. BPEV-QC and PvEV-JD shared high homology with other same reported isolates, respectively, kinship nearly, and shared low homology with other endogenous RNA virus isolates, kinship far away.(4) Analysis of BYMV-JD' enzyme loci known that the characteristics of the enzyme loci were basically identical with other members of the genus PVY. Analysis of BYMV-JD' genome structure known it had motifs of GHSG, CCC, PTK, GAVGSGKS and GQCG. Analysis of the homology of the nucleotide and amino acid sequence and phylogenetic tree known BYMV-JD shared highest homology with AR87C, kinship nearly, and shared lowest homology with 90-2, kinship far away.
