| The melanins from mammalian epidermis are responsible for the hair colors by the number and distribution. TGFβ is one of the multifunctional cytokine, which plays important roles in apoptosis, cell differentiation and cell activities. TGFβ3 secreted by melanocytes can affect the absorption of melanin and melanin synthesis. However, whether the exogenous TGFβ3 regulates melanogenesis is poorly understood. In this study, alpaca melanocytes with different doses of supplementary of TGFβ3 in medium were used to analyze the phenotype, color-related genes MITF, TYR, TYRP2 expression and melanin production, which will reveal the effect of TGFβ3 on melanogenesis in alpaca melanocytes.The main contents and results are as follows:1. Followed the supplementary of TGFβ3 with different doses (6.25ng/ml,12.5ng/ml,25ng/ml, 50ng/ml) in medium of 56h, the alpaca melanocytes appeared obvious poles projections, compared with the control group.2. Add alpaca melanoma cell proliferation TGFβ3 on:TGFβ3 of melanoma cells to maintain long-term effects of cell numbers, but no dose lazy by nature.3. The quantative real-time results showed that mRNA expression of MITF were 0.513,0.524,0.535, 0.314 fold changes, mRNA expression of TYR were 0.490,0.712,0.562,0.575 fold changes, mRNA expression of TYRP2 were 0.408,0.403,0.458,0.509 fold changes, respectively in melanocytes with dfferent doses of TGFβ3.4. The Western blotting results showed that protein expression of MITF were 0.780,0.830,0.790, 0.700 fold changes, protein expression of TYR were 0.677,0.850,0.640,0.620 fold changes, protein expression of TYRP2 were 0.870,0.900,0.700,0.600 fold changes, respectively in melanocytes with dfferent doses of TGFβ3.5. Spectrophotometry results showed that melanin production in melanocytes with different doses of TGFβ3 were 0.736,0.800,0.750,0.665 fold changes, compared with the control group.Conclusions:TGFβ3 affect the expression of MITF, TYR and TYRP2 and the regulation of the melanin production. However, it may promote the transferring of melanins from melanocytes to keratinocytes around by the processes. |
