The purpose of this study is to explain the effect of dietary fiber level on the interaction between M cells and antigens,and study the effects of different fiber levels on the growth performance,slaughter performance,muscle quality and gastrointestinal and immune organ development influences.And to provide theoretical basis for the regulation of intestinal mucosal immunity by other animal and human factors,and to support nutrient as an immunomodulator.1.Effects of Dietary Fiber Levels on Growing Meat RabbitsA total of 120 rabbits were randomly divided into three groups,each with 40 replicates,one for each trial,and fed with low(LF,CF14.32%),(MF,CF18.46%),high(HF,CF 22.37%)of three different fiber levels of the test diet.Pre-trial period 7 d,is the trial period 20 d.Test results are as follows:(P = 0.001 2,P = 0.021 9 and P = 0.000 1)were significantly affected by different dietary fiber levels(ADG),mean daily feed intake(ADFI)and feed /,ADG highest or F / G minimum appears in LF group.(P = 0.018 0,P = 0.036 6 and P = 0.034 1)had significant differences in the effects of different fiber levels on the live weight of the pre-slaughter,half-bore body weight and full-bore carcass There was no significant difference between the slaughter rate and the full slaughter rate(P> 0.05).The dietary fiber levels had no significant effect on the pH,the brightness value(L *),the red value(a *)and the yard value(b *)of the musculoskeletal muscle(P> 0.05).There was a significant difference in the relative weight(P= 0.011 5,P = 0.022 0 and P = 0.031 1,P = 0.031 1)between the different dietary fiber levels on the stomach,cecum and its contents,and the HF diets were significantly higher LF diet group(P <0.05).The dietary fiber levels had significant effects on the weight of thymus,spleen and liver(P = 0.005 2,P = 0.006 8 and P = 0.033 8).With the increase of dietary fiber level,the weight of immune organs increased significantly Big.In addition,the spleen index reached the maximum in the MF group,but there was no significant difference between the LF group and the LF group(P> 0.05).Effects of Dietary Fiber Levels on the Interaction of Intestinal M Cells and Antigens in RabbitsEight rabbits were randomly selected from the three diets group,and the rats were randomly selected from 8 rabbits.The cells were cultured in vitro and seeded with the same amount of XL-Gold containing green fluorescent protein.Escherichia coli(indicator bacteria)were cultured for 10 min,30 min and 1 h respectively.The paraffin sections were made by tissue immobilization.The same number of indicator bacteria were inoculated at the end of 10 min,30 min and 1 h respectively.The paraffin sections were made after three different time points.The The expression of N cells in the lymph nodes was measured by using the anti-vimentin antibody.The number of M cells in the lymphoid tissue was measured and the levels of M cells On the carrying capacity of a particular pathogen.The results are as follows:2.2 In vitro tests(P <0.05),but there was a significant effect on the number of M cells in the tissue at 30min(P> 0.05)(P> 0.05).There was no significant difference in the number of M cells in the culture group(P> 0.05).2.3 In vivo testsThe expression levels of RANK,RANK-L and OPG genes in HF group and LF group were not significantly different(P> 0.05).The expression level of RANK and OPG gene in vitro was 10 min,30min(P <0.05),and there was no significant effect on the expression of RANK-L gene(P> 0.05).There was no significant effect on the expression of RANK,RANK-L and OPG protein(P> 0.05).There was a significant effect on the number of M cells contained in the larvae of Eupatorium adenocarcinoma at 30 min and 1 h(P <0.05)(P <0.05).The appropriate fiber level was 18.46% in the diet of weaning(35 days)to 65 days old.The high fiber level diet could cause M cell morphology and quantity change,so we thought that the regulation diet The level of fiber can affect the transport capacity of M cells to the indicator bacteria.
