Bats are of rich species of mammals in the world,there are nearly 1200 kinds of different species.They can distribute in except for the polar regions of the world.Bats are also important carriers and can carry at least 170 species of viruses.In 2012,the researchers found H17N10 in bats.But it is not clear that Batsare hosts of influenza virus.Bat influenza A-like virus and other influenza A-like virus are very similarin genome structure,there are also many of the different places.We got genome information of H17N10 through a new generation sequencing means,But we can not rescuebat influenza A-like virus H17N10.In zhis study we successfully rescued recombinant bat influenza A-like virus by The reverse genetic operating system,the recombinant bat influenza A-like virus was named cH9cN2 / H17,cH1cN1 / H17.To study the cH9cN2 / H17,cH1cN1 / H17 growth rule in MDCK,DF1 cells,we take the virus of MOI,0.001 the amount in the MDCK,DF1 cells.We received the samples in 12 h,24h,36 h,48h,after it we examinate50% tissue cullure infective dose(TCID50)of the virus.The results show that50%tissue cullure infective dose(TCID50)of bat influenza A-like virus cH9N2 / H17,cH1N1 / H17 reached the highest in MDCK cells and DF1 cell in 36 h.In order to study the biological characteristics of the cH9cN2 / H17,cH1cN1 / H17 in mice,106TCID50 cH9N2 / H17,cH1N1 / H17 infect in mice.The results show thatcH9N2 / H17,cH1N1 / H17 do not cause weight loss and other anomalies in mice.But after three days,five days,we detected the virusin the lungs of killed mice.After15 days we can detect the serum antibodies in mice.In conclusion we successfully established the Reverse Genetic System of bat influenza A-like virus,provided certain reference basis on bats recombinant inflienza virus breedingin MDCKcellsandDF1 cells.We also made a research on the biological characteristics of the virus in mice.These studiesprovided support Forbat influenza A-like virus cH9N2 / H17,cH1N1 / H17 pathogenicity on mammals.
