According to the published sequence of Streptococcus suis in GenBank. this study designed specific primers for SAO protein gene and named SAO-D, the amplification of stripe size to 400 bp. In order to succeed to distinguish the appearance of protein gene different variants and identification of SAO protein gene is missing, According to the SAO protein design specific primers, and named SAO-FL. In different types of SAO protein gene, the size of the target fragment is different:SAO-L is about 2500bp, SAO-M is about 2000bp, SAO-S is about 1700bp and the gene deleted strain is about 300bp.Our laboratory separationed and saved a total of 318 strains of streptococcus suis, 269 successfully identified serotype strains, were covered by the serotype of 25. All type serotype 16 strain detected SAO protein gene, and in the serotype 2,8,9,15,28, 31 are some strains can't detect SAO protein gene. Successfully detected SAO protein gene of the strains in 243 strains, accounting for 90.3% of the total amount of testing, SAO protein gene deletion strains 26 strains, accounting for9.7% of the total volume. Detected in all successful protein gene of streptococcus suis, SAO-M protein gene main popular forms of SAO, a total of 192 strains, accounted for 79% of the check, SAO-L, a total of 18 strains, the detection rate was 7.4%, SAO-S detection rate, at least 7 strains, the detection rate of 2.9%. some samples on SAO-FL parting, failure to a total of 26 strains, accounting for 10.7% of the total volume. Therefore. SAO protein is widespread in the streptococcus suis, and in many variants of SAO protein, SAO-M is the most common variants. With SAO protein recombinant protein as streptococcus suis antigen protein subunit vaccine, will gain a larger range of protective.
