Resveratrol as a natural product extracts, studies found that it not only palys important roles in anti-tumor, anti-cardiovascular disease, anti-inflammatory, antibacterial and antiviral, anti-aging, but also has great effects in the reproductive system. In the present study, we used in vitro cell model combing with in vivo animal study and experimental study of the antioxidant combined in vivo animal to explore its role in anti-oxidative damage.In vitro experiments:a culture system of primary porcine Ley dig cell was established, in which cells were isolated from piglets with one month age. Hydrogen peroxide was used to reduce the oxdative damage. The data showed that a dose of 150 ?M or higher could induce the oxdative damage which resulted the abnormal cell morphology, inhibition of cell proliferation, significantly increasing the cell apoptosis, decreasing theproduction of antioxidant SOD, and as well the cells'ability to secrete testosterone (T). Low doses of resveratrol could promote the proliferation of the primary Leydig cells, the treatment with a concentration of 0.2?M obtained a ideal effect. In the oxdatice damage cell model, treatment with resveratrol, in partially, could protect the cells from oxidative damage caused by hydrogen peroxide showing as reducing the inhibition of cell proliferation cuased by hydrogen peroxide, increasing antioxidant SOD generation in oxidative damage cells, meanwhile, improving the ability of testosterone secretion and decreasing the apoptosis. The data obtained from in vitro study may provide the basis for following animal work in vivo.In vivo animal experiments:we use unmature male mice to establish the spermatogenesis in mice which was incuded cyclophosphamide. In this animal model, reproductive toxicity is obvious:the indexes of testis and epididymis were significantly decreased; total number of sperms was declined and the abnormal rate of sperm was increased; In the mouse sera, the levesl of anti-oxdatants SOD and GSH-Px showd no change, and the concentration of the testosterone was increased when compare to the model group. The pathological damage in tesits was also reduced. Moreover, no effects in preproductive performance were observed when only using the resveratrol with the above mentioned dose.All these results suggest that resveratrol showed a therapeutic effect on anti-oxidative damage, but its specific mechanism needs to be done in the future work. As its effects both in in vitro cell system and in vivo animal model, these data may supply a reference for resveratrol used an important antioxidant drug or as a feed additive in animal production.
