Goose parvovirus is the member of Parvovirinae,it can infect geese and Mus covy ducklings to cause a high morbility and morbidity disease.Since 2015,there are reports that Cherry Valley duck farms break out a disease of short beak and dwarfism syndrome(SBDS)in mainland China.Through isolation and identification by Chen et al.,it is confirmed that this virus belongs to a novel Goose Parvovir us(NGPV).The aim of our study is to carry out an epidemiology investigation fo r Dezhou that located in Shandong province,meanwhile we carry out an exper-ime nt of sequencing the complete genome of 1 separated strains and the homologous analysis and phylogenetic tree analysis.1.The epidemic investigation of the goose parvovirus in Cherry Valley duck flocks of Dezhou,Shandong provinceAccording to the data of the GeneBank published already,we design the detection primers of GPV using Primer 5.0.We carry out the detection with extracting DNA from the heart,liver,kidney,spleen,lung,thymus pancreas,and bursa of Fabricius,brain,bile 10 viscera tissues total of 400 samples of the epidemic materials that come from 5 flocks,we analyze the the positive rate of every flock and statistics the rate of organs.Results showing that with the method the highest and the lowest positive rate is 100%and 62.5%,and the average positive rate is 82.5%;and the highest and the lowest positive rate of every viscera is 82.5%(Heart)and 15%(Lung)without brain and the average positive rate is 45%.The results of epidemic investigation show that NGPV prevelants in Dezhou and causes big loss to Cherry Valley duck flocks.2.The complete genome sequencing and gene evolution analysis of DZ-01 strainAccording to 12 strains of GPV and 2 strains of NGPV sequences from GeneBank published already,we designed 5 NGPV sequence pairs for complete genome amplification using Primer 5.0 software,synthesizing by Shanghai Sangon biological engineering company.The homology analysis results show that DZ-01 have a homology rate of 92.7?97.2%and 99.3?99.7%with QH15 and sdlc01,but just 85.2%with a MDPV of SAAS-SHNH strain.The result of evolutionary tree analysis shows that the DZ-01 strain belongs to the branch of GPV,it is in the same small branch with sdlc01 and QH15;We also give a homology analysis to VP3 gene,the result indicates that it is the lowest homology with GDaGPV,but has the highest homology of 98.5%with 82-032v,DZ-01 strain is in a high homology with sdlc01 and QH15 of 99.7%and 100%.We also conduct a homology analysis for VP3 protein,finding that both DZ-01 strain has the lowest homology rate of 96.6%with GDaGPV strain,and DZ-01 have the highest homology rate of 98.7%with B strain,the homology rate with sdlc01 and QH15 is 100%and 99.6%.Evolutionary tree analysis showed that DZ-01 and sdlc01 and QH15 are on the same branch,but having a far relationship with classical GPVs.
