Tomato(Solanum lycopersicum L) originated in South America, in our county it has many years of cultivation history, now it has become one of the important economic crop in C hina. In southern China, High Temperature in Summer or greenhouse temperature seriously affect the normal growth and development of tomato, Heat Stress has beco me a major cause of tomato production. We studied 150 tomato inbred lines to heat on physiological response, provide a theoretical basis and technical support for the identification and screening of high temperature resistant varieties, formally established the methods of tomato resistant identification system. Using SSR molecular marker technology with cluster analysis, we analysed genetic diversity of 150 tomato material. Through tomato heat shock protein, heat shock transcription factor gene bioinformatics analysis to study differences in gene expression of heat shock protein at heat stress.(1)In this experiment, 150 tomato inbred lines were under high temperature treatment, detected the change of malondialdehyde and antioxidant enzyme activity and proline content, finally get resistant and sensitive variety to high temperatures.We established a new resistant screening system based on chlorophyll, conductivity and proline. Successfully screened out 15 heat-resistant material, 12 heat-sensitive material parts.(2)We use tomato DNA as templates, 13 pairs of SSR primers that amplified clear bands specific were screened from 83 pairs. With cluster analysis using UPGMA method, draw tomato SSR markers genetic similarity dendrogram. On the similarity coefficient level of 0.65, the resources can be divided into two types. They were common large tomatoes and small cherry tomatoes.(3) Through the biological information analysis to study tomato and Arabidopsis heat shock protein gene and heat shock transcription factor genes and construct phylogenetic trees. We get 166 heat shock protein genes in tomato, contain 7 HSP 90,19 HSP 70,7 HSP 100, 12 HSP 40,21 points HSP 60,24 heat shock transc ription factor genes. By screening the SNP information that closely related to the tomato heat shock protein gene, initially clear the species and distribution of SNPs which located on heat shock protein gene, and we know that the most SNPs located in non-coding region. By Q-PCR analysis of heat shock protein gene expression levels under heat stress.
