This thesis researched the introduction of Hirudinaria manillensis and growth rule, overwintering and reproduction characteristics of introduction Hirudinaria manillensis in Guizhou province. and explored the hirudin activities characteristics of introduction Hirudinaria manillensis in different developmental periods and hunger situation, aims to provide a reference for the Hirudinaria manillensis farmed. The study results and conclutions were as follows:1.The mode of introduction transportation of Hirudinaria manillensis and domesticate characteristic of introduction Hirudinaria manillensis has been studied.The results showed that:(1) the survival rate was 99.55% by the means of dry ice transportation.(2) The differences were not significant of growth rate and survival rate of introduction Hirudinaria manillensis when the stocking density in 0.5 kg/m2, 1kg/m2 and 1.5 kg/m2 for density group, the growth rate and survival rate significantly reduce when the stocking density more than 1.5 kg/m2.(3) The growth rate of cluster feeding by Sun-shade net more 28.40 percent than traditional way of feeding directly.The results of the experiment showed that the Hirudinaria manillensis can make a successful introduced by means of dry ice transportation. The stocking density should be lower than 1.5 kg/m2 is appropriate in the process of cement pool breeding.2. The growth rule of introduction Hirudinaria manillensis were studied by the the means of biological statistics and biochemical. The results showed that:(1) the taking coefficient and feed coefficient of Hirudinaria manillensis were gradually decreased with the increase of the weight, the period of averaging rapid metabolic were gradually extension with the increasing of the weight, the growth rate of were decreased with the increasing of the weight. The taking coefficient has significant differences in the stage of less than 1g, 1~4g, 4~10g, and more than 10 g, the feedingIV periods of Hirudinaria manillensis also has differences, and we could grading feeding Hirudinaria manillensis according to it.(2) the trypsin activities were highest in subadult period, with the growth and development, the pepsin activities were increased gradually. Different digestive enzyme were different in different periods after or before feeding.3.The influence of the different soil matrix, overwintering density and overwintering equipment to Hirudinaria manillensis during overwintering were studied. The results showed that the yellow soil is better than paddy soil as a winter matrix, the winter survival rate of different density group were low, the survival rate of slope cement pond is higher than plastic case, analysis that the reason maybe due to the bottom hydrops, lead to gas exchange was influenced, and is bad for temperature stability, lead to death. and then death Hirudinaria manillensis breed bacteria, lead to soil destruction, cause secondary infection causes the cluster group death, made the survival rate is low. The results of the experiment showed that the Hirudinaria manillensis could succeed during overwintering by the yellow soil as a winter matrix in Guizhou natural environment.4. The reproduction characteristics of introduction Hirudinaria manillensis were studied. The results showed that the mating rate were 54.7±1.2%, the survival rate and the weight-loss ratio after the cocoon production were 91.67±2.36%, 26.91±0.93%,respectively. the spawning cocoon number were 3.8±0.61 gold/article, the average cocoons spawned quality were 1.16±0.25 g, and the cocoons spawned hatchability were 90.67±6.11%. The body weight and body length of first young Hirudinaria manillensis larva. The results of the experiment showed that the Hirudinaria manillensis would succeed reproduction in Guizhou natural environment, in addition,the rate of weight loss is bigger after the cocoon production, should strengthen nutrition supply.5. The hirudin activities of introduction Hirudinaria manillensis of different development period, different starvation time, different apparent symptoms of death,and different varieties of leech were studied by the the method of thrombin drop point.The research showed that: the unit mass activities of hirudin were gradually decreasedwith the growth and development, and the activity of hirudin reach 73.24±3.65ATU/g was highest when the larva, and when the adult was least, which was50.46±2.88ATU/g. The unit mass hirudin activities of introduction Hirudinaria manillensis was decreased after feeding, and was significantly increase before feeding again, and then, the hirudin activities has a falling trend. The hirudin activities of different apparent symptoms of death was not significant, and lower than the normal group. The hirudin activities of different varieties of leech was Hirudinaria manillensis > Hirudonipponia Whitman > Whitmania acarnulata Whitman >Whitmania pigra Whitman, and the hirudin activities were 50.39±2.55 ATU/g ?43.28±3.23 ATU/g?9.38±1.63 ATU/g?3.13±1.07 ATU/g, respectively.
