Galls are extremely common in nature,which are the peculiar product of the interaction between insects and plants.The gallwasp was one of the main groups of gallmakers.Gallwasp lives in the completely closed galls,intercepting host plant nutrition and impairing plant seriously.It is difficult to control on account of highly hiding.Therefore,building accurate and efficient identification system of gallwasp was necessary.As a new species identification technology,DNA barcode technology has been widely concerned.DNA barcoding technology was applied to the gallwasp of China for the first time.Basing on molecular markers of CO??CO??28SrDNA and combind database of the three genes(CO?+CO?+28SrDNA),we reconstructed the phylogenetic relationships of 4 genus 22 species belonging to Cynipidae through Neighbor Joining,Maximum likelihood and Minimum evolution methods.The results were as follows:1.The result of base analysis show that,the contents of A+T higher than G+C to CO??CO?and dataset(CO?+CO?+28SrDNA),the contents of A+T lower than G+C to 28 SrDNA.The proportation of Variable sites to CO?higher than other data,having the advantages to differentiate sibling species.ratio of transition to transversion for CO? was 0.69,ratio of transition to transversion for CO? was0.64,show the characteristics of high mutation by the mitochondria gene.ratio of transition to transversion for 28 was 1.85,show high conservation by the ribosomal gene.Ratio of transition to transversion for database was 0.8.2.The result of genetic distance show that,intraspecific variation range of genetic distance to CO?is 0-0.014,interspecific variation range of genetic distance is 0.017-0.266.there was DNA barcoding gap exist.We can identify species accurately relying on the DNA barcodes.The other data has an overlapping region in intraspecificd and interspecific genetic distance.3.The result of Phylogenetic signal show that,CO?,CO? and dataset(CO?+CO? +28SrDNA)have a good linear dependence between p distance and R.The transition and transversion ratio of all data didn't reach saturation,and they can be used for phylogenetic analysis.4.The result of phylogenetic tree show that,the monophyly of Synergus was supported,but reject monophyly for Saphonecrus.Synergini and Cynipini was sister relationship.The Maximum Likelihood tree was the most efficient ways to reconstruct the phylogeny tree of Cynipidae.The phylogeny tree based on the dataset(CO?+CO?+28SrDNA)has more clearly Phylogenetic relationships.Therefore,various methods of classification should be integrated inorder to achieve accurate identification of species.
