This paper was aim to detect the expression of MSTN/Smad signaling pathway gene(MSTN, Smad2, Smad3, Smad4, Smad7, TGFβR Ⅰ, TGFβR Ⅱ) at birth and 5 months in different tissues(Heart, liver, spleen, lung, kidney, intestine, femoral biceps, triceps shares,semitendinosus, semimembranosus, longissimus muscle and fat) and 1-5 minth in Bashiby sheep By RT-PCR,and Analysis the MSTN/Smad signaling pathway gene expression level correlation with their growth Characters in different months,exploring the expression of MSTN gene at different tissues, and different developmental stages in Bashiby sheep; also studied expression level of MSTN gene correlation with DNA methylation, provide the basis for molecular biology genetics and breeding for Bashiby sheep.1. The study MSTN/Smad signaling pathway gene expression in all tissuesesAt birth: The MSTN gene expression in fat was significantly higher than other tissues(P<0.05), spleen, femoral biceps, semitendinosus were significantly higher than other tissues except intestine and fat(P <0.05); The Smad2 gene expression in kidney and fat were significantly higher than other tissues(P <0.05); The Smad3 gene expression in visceral tissues was significantly higher than muscle tissues except longissimus muscle(P <0.05); The Smad4 gene expression in visceral tissues was significantly higher than muscle tissues except lung,semitendinosus, longissimus muscle and fat(P <0.05); The Smad7 gene expression in liver, spleen, lung, intestine,fat were significantly different with kidney,femoral biceps, triceps shares, semitendinosus, semimembranosus, longissimus muscle; The TGFβR Ⅰ gene expression in spleen, kidney, intestine were significantly higher than other tissues(P<0.05);The TGFβR Ⅱ gene expression in Heart,lung, kidney,fat were significantly higher than other tissues(P <0.05);At 5 month: The MSTN gene expression in fat was significantly higher than other tissues(P <0.05);The Smad2 gene expression in fat were significantly higher than other tissues(P<0.05), There was no expression in the liver, triceps shares and longissimus muscle; The Smad3 gene expression in lung was significantly higher than semimembranosus(P<0.05),There was no expression in triceps shares; The Smad4 gene expression in spleen and kidney tissues was significantly higher than in riceps shares, semimembranosus,longissimus muscle,fat(P <0.05); There was no expression in lung, femoral biceps, semitendinosus; There was no significantly with tissues of the Smad7 gene expression; The TGFβR Ⅰ gene expression in fat was significantly higher than other tissues(P <0.05);The TGFβRⅡ gene expression in liver was significantly higher than intestine(P <0.05);2. The study MSTN/Smad signaling pathway gene expression in different month Muscle: The MSTN gene expression at 1 month was significantly higher than 2 month, 3month,4 month and 5 month(P<0.05), at birth was significantly lower than 2 month, 3month,4 month and 5 month;The Smad2 gene expression at 5 month was significantly higher than other months( P<0.05); The Smad3 and Smad7 gene expression at 4 month was significantly higher than other months(P<0.05); There was no significantly of the Smad4 gene expression(P>0.05); The TGFβRⅠgene expression at 2 month and 3 month was significantly higher than other months(P<0.05); The TGFβRⅡgene expression at 2 month,3month and 4 month was significantly higher than at birth, 1 month and 5 month(P<0.05).Fat: The MSTN gene expression at 5 month was significantly higher than other months(P<0.05), 3 month was significantly lower than at birth, 1 month and 4 month; The Smad2,Smad4, Smad7 gene expression at 4 month was significantly higher than other months(P<0.05); The Smad3 gene expression at 3 month was significantly higher than other at birth, 1 month,2 month and 5 month(P<0.05),4 month was significantly higher than at birth,1 month,2 month and 5 month(P<0.05); The TGFβRⅠgene expression at 3month,4 month was significantly higher than other months(P<0.05); The TGFβRⅡgene expression at 4month was significantly higher than other months(P<0.05).3. MSTN/Smad signaling pathway gene and growth traits associated with each gene analysisMuscle : The expression of MSTN gene and body weight was a significant negative correlation(r =-0.864, P <0.01), Smad3 genes and body weight was significantly negative correlation(r =-0.522, P <0.01), with the cannon circumference was a significant negative correlation(r =-0.479, P <0.05), TGFβRⅠ gene and body length was a significant negative correlation(r =-0.461, P <0.05); MSTN gene with Smad3 gene was a significant negative correlation(r =-0.421, P <0.05), and with TGFβR Ⅰ gene was negative correlation(r =-0.342).Fat: The expression of Smad2 gene,Smad3 gene and body length was significantly positive correlation(r = 0.617, r = 0.588, P <0.05), Smad7,TGFβRⅡ gene and body length were significantly correlated(r = 0.640, r = 0.632, P <0.01); with TGFβR Ⅰ gene was negative correlation(r =-0.334).4. MSTN gene DNA methylation correlation analysisBiceps femoris, femoral triceps, semitendinosus, semimembranosus, methylation probability longissimus muscle and the fat tail of 74.2%、74.2%、83.2%、83.7%、82.1% and25.3%。. And gene expression MSTN was a significant correlation(r =-0.48, p <0.05). DNA methylation has a certain effect on carcass traits. |
