Wheat is one of the major crops in the world. Studing the genetic basis of yield traits and exploring yield gene are important to improve wheat production. However, there are few reports on cloning of yield gene in wheat. In this study, we cloned a seed development-related gene family in wheat, named Tazmhl22. It inclued four homologous genes at least, and encodes a novel class of proline-rich proteins (PRP). We cloned four genes of Tazmh122 whose coding regions have similar repeat sequences, and the four Tazmh122 are significantly distinct from the five classes of plant PRPs described previously. Semi-quantitative RT-PCR showed that Tazmh122 wis highly expressed in reproductive organs, and the expression levels were reduced according to the developmental stages of seed. Using 136 recombinant inbred lines derived from Nanda2419 and Wangshuibai, linkage analysis showed that Tazmh122.3 located within the interval Xmag2271?Xwmc398 on chromosome 6BS. One hundred twenty-five Chinese mini core collection were genotyped and association analysis revealed that Tazmhl22.3 was significant associated with grain weight.
