Job's-tears is one of the world's most pop? lar medicine edible plants of annual or perennial C4 Poaceae,and it has a reputation as “king of the world graminious”.European even regard Job's-tears as “the grain of world health”.Their principal effective constituent is neutral oil content of Job's-tears used for anti-cancer injection,which as the first Chinese medicine was approved by FDA for Clinic ? trials in 2015.As a small species in plants,Job's-tears has no genomic information in the NCBI database so far.This has been aggravated mainly to the limited availability of germplasm resources and low genetic polymorphism in Job's-tears.However,the availability of RAPD markers provides opportunity to reveal genetic polymorphism among numerous alleles,especially in the associated-markers with embryo oil content in wide-diversity of Job's-tears.These data will serve as an important resource to assist breeding programmes in Job's-tears.In the present study,we sough to identify functional markers associated with the embryo oil content and find out excellent varieties of Job's-tears.In the first place,we collected twenty-three kinds of Barley rice germplasm,named as LG001~LG023,which were furnished by Institute of Bioengineering,Zhejiang Sci-tech University(LG010 comes from Shanxi,LG011 comes from Liaoning).To isolate the genomic DNA among twenty-three kinds of Barley rice germplasm,we adopted modified CTAB process.All DNA samples which were qualified by Nano Drop 2000 and 2% agarose electrophoresis,sho? ld be diluted to 10ng/? l,-20? storage for reserve.The sequence informations of Job's-tears(small species)RAPD primers come from relevant documents about RAPD's primer selection of Poaceae(Rice and Wheat,genetic closely related to Job's-tears).34 RAPD primers were screened out from 256 RAPD primers with stable,clear amplification and synthesized by Shanghai sangon.Statistical analysis is based on the RAPD amplification products,there is a stripe down to 1 and no stripe down to 0.In addition,it sho?ld help to set up a database on which to base statistical stripes.Based on RAPD fingerprinting,the 23 germplasm resouces were clustered to 5 groups using 0.75 coefficient as a standard via UPGMA method,which were in accordance with the geographical resource and germplasmic genealogy.Using Mathematical Methods of Physics,Contents of 23 kinds of Job's-tears neutral oil was examined,LG013 occupies the 8.99%-strong flock(average 3.47% of twenty-three germplasm),the highest level among all the 23 kinds of Job's-tears species.LG013 is the best variety among the twenty-three germplasm,it has the largest grain long 11.222 mm,largest grain wide 10.148 mm,largest grain height 8.194 mm,the biggest embryo oil content is 0.00875 g,about 8.99373%-strong flock.Sim?ltaneously,We found that LG013 has the specific band in the 402 th among 405 alleles by S44 RAPD primer,the molec?lar weight is about 1050 bp.Afer a series of cloning,DNA sequencing,Primers(F:TCTGGTGAGGGGAGTCACA,R:TCTG GTGAGGCATGGTGT)were designed out.The identification reveals that LG013 has a unique band in 600 bp not band by anybody else,which may be closely related to embryo Oil content of Job'-tears.It's demonstrated that the 600 bp unique band can be used for the variety identification of LG013 germplasm and application for protection of intellectual property.
