As the largest digestive organs with most function, liver can convert and store up the harmful substances of body metabolism and heterologous substances, This makes liver vulnerable to these toxic substances, causing a variety of acute and chronic liver damage and even causing liver cell necrosis or apoptosis. At present, There are a lot of liver-protective drugs on the market, but it have significant side effects and the price is so high, those caused limitation in clinical application. Therefore, developing natural medicine with good liver-protective effect and little side-effects is first concerned problem. Pine pollen is a traditional medicine and edible pollen. Because it is rich in nutrients, it has a variety of biological effects, including liver-protective function. Pine pollen polysaccharides is a biologically active macromolecules has been shown to have a variety of biological functions, such as antioxidant and immune regulation. But its liver-protective effect is rarely reported. Therefore, this study established two chemical liver injury model to study the protective effect of pine pollen polysaccharides on liver injury and its part of mechanisms from the whole and the molecular level. The main results were as follows:(1)In this study, water extraction and alcohol precipitation method was used to obtain pine pollen polysaccharide, and then the TPPPS were disposed by decolorizing and protein processing in order to get the high purity polysaccharide. The polysaccharide content was 73.46%; Detected by infrared spectroscopy for polysaccharide characteristic absorption peaks. This lay the foundation for the next structure of pine pollen polysaccharide used for further analysis. By measuring the pine pollen polysaccharides on DPPH ?, ? OH, O2- scavenging and reducing power, it showed pine pollen polysaccharides has good antioxidant activity in vitro, with a positive correlation between the concentration of the polysaccharide.(2) By establishing CCl4 acute liver injury model in rats, we investigates different doses of pine pollen polysaccharide intervention effects on liver injury. Results demonstrate pine pollen polysaccharide can effectively reduce serum enzymes related to liver function, vigor of ALT,AST, ALP and liver index of levels in liver damage rats; Using HE staining of paraffin sections of liver under an optical microscope demonstrated the pine pollen polysaccharides played a good protective effect on liver cell structure; When we detected antioxidant activity on rats, we find pine pollen polysaccharide can effectively increase SOD, GSH-Px in serum and liver and reduce MDA content, it proved pine pollen polysaccharide played a good protective effect on pathway for liver injury by resistance oxidative stress.(3) Through the establishment of chronic alcoholic liver injury in mouse model, we investigates different doses of mountain Tai’s pine pollen polysaccharide intervention in chronic alcoholic liver injury. Results demonstrate pine pollen polysaccharide can effectively reduce serum enzymes related to liver function, vigor of ALT,AST, ALP and liver index of levels in liver damage rats. We use electron microscopy observe ultrastructure of liver cells and found that pine pollen polysaccharide play a protective role in endoplasmic reticulum normal structure. We use Western-blotting technique to quantitative analysis expression of endoplasmic reticulum stress-related protein GRP78 and CHOP protein. It proved that pine pollen polysaccharide can play a protective role on endoplasmic reticulum stress.In conclusion, TPPPS has the apparent protective effect to acute CCl4 liver damage and chronic alcoholic liver injury, its mechanism is related to improving antioxidant activity, scavenging free redical and inhibiting endoplasmic reticulum stress.
