Based on agricultural solid waste straw streptomyces is a degradation product of polysaccharide extraction,separation,purification and structure analysis,straw degradation waste composting improvement of saline alkali soil,soil physical and chemical properties of the to achieve the purpose of straw resource utilization.Experiments in crop straw vaccination has been screened for straw degradation optimization strain continuous fermentation experiment was carried out and concentrated supernatant extraction,separation purification purified polysaccharide(molecular weight range of polysaccharide component),by physical and chemical methods of analysis of polysaccharide primary structure: The molecular weight,functional group,the composition of the simple sugars and the surface characteristics were characterized.Collection of fermented residue,dry soil covering returning for research on improvement of saline alkali soil,through analysis on soil physical and chemical properties and plant growth status,indicating that waste composting for saline alkali soil improvement effect.The experimental results are as follows:First,single open type continuous fermentation conditions of operation of the device is:actual fermentation volume of 5L of 70%;operating temperature at 35 ?;pH control in8.00~8.55;operating pressure of 0.1MPa;microorganism 1:1:1,4% of the amount of bacterium inoculated into the feed tank;feed tank stirring speed control in 300 r / min;peristaltic pump parameters: velocity of 0.4ml/min.Fermentor agitation speed control in 300r/ min,pH8.00~8.55,temperature of 35 ?;the tank material pump into the fermentor to be80%,stop at 12 h reaction.At the same time,the fermentation tank material to 0.4ml/min speed into the storage tank.Compared with shake flask fermentation,the sugar yield of continuous fermentation was increased by 14.63 ~100.41 mg/L.Secondly,polysaccharide extraction by water extraction and alcohol precipitation method,through the comparison of three kinds of protein removal.After savage method after removing protein,protein content is 5 ? g / L,is reduced by two orders of magnitude,compared with activated carbon and hydrogen peroxide bleaching effect,hydrogen peroxide bleaching polysaccharide loss rate of decolorization effect good.Purification of polysaccharides by sephadexg100,two kinds of molecular weight components,FFSP1 is 7.2× 104 Da,FFSP2 is 3.3 × 105 Da.Repeated solution frozen centrifugation and UV scan showed the purified proteins and polysaccharides,nucleic acids decreased and its structure with general carbohydrate with functional groups,an acidic polysaccharide,with beta D grape adjoin furanose characteristics,environmental scanning electron microscopy analysis results showed that the polysaccharide samples showed patchy or clastic accumulation and smooth surface.The results of GC-MS analysis showed that was composed of rhamnose,xylose,mannose,Arabia sugar,glucose,galactose.Again,comparing the two types of returning showed that straw residue compost and soilpH reduced 0.30~0.85 units and returning straw to field directly,the saline alkali soil pH reduced the 0.05~0.49 units,and that residual straw compost soil organic matter and available nutrient content than the straw returning to soil improvement effect significantly,and wheat seedlings grow well,visible straw residue compost incorporation of saline alkali land having improved effect.With increase of the dosage of composting with straw compost to 1:12(mass ratio)covering soil,more conducive to the improvement of wheat growth and saline alkali soil.In order to further study the migration of ions in the soil of different texture transformation rules,simulated soil environment,design of soil leaching device,including organic glass material leaching device experiment box,pebble bearing layer,anti seepage wall,column experiments overflow,infiltration filtrate water inlet and outlet valves and stents.Device is designed for the length,width and height,respectively,is 63 cm,21cm,52 cm cuboid soil column structure,column body is divided into three sections,from top to bottom set three layer sampling port can realize online monitoring of soil nutrient leaching and migration characteristics of,to create conditions for the subsequent experiments.Through the comprehensive development and utilization of straw,to achieve the purpose of agricultural solid waste resource utilization.
