Production Performance, Blood Lipid Index And Their Association With Lipid Candidate Gene Expression In Chinese Indigenous Wuhua Chicken

In Chinese indigenous broiler breeds, the production performance and meat quality showed high difference and also influenced by sex and slaughter age. Intramuscular fat(IMF), one of the important factors of meat quality, is regulated by many genes during deposition. In order to measure meat quality and the expression of genes affecting meat quality, 80 hens and 80 roosters were reared under the same conditions, and 10 of each were randomly selected during day 90, 120, 150,180 for slaughter. IMF, IMP and lipid content were determinded in thigh and breast muscle. Expression level of L-FABP, FAS,Apo B and PPAR ? was determined and their correlation with IMF was analyzed. The results were as follows:1. Meat performance of Wuhua chickensLive weight, carcass weight, half-eviscerated weight, eviscerated weight, gizzard weight and chest muscle weight, leg muscle weight, heart weight, liver weight of roosters have no significant change in day 90 and 120, while that of day 150 are bigger than that of day 180(P<0.01). Breast muscle weight, leg muscle weight, heart weight, and liver weight of hens have a significant increase after day 120(P<0.05). The average weight of abdominal fat of rooster in day 90 and 120 are 3.85 g and 5.85.In day 150, 180.of rooster and day 90,120 of hens, there are no abdominal fat visible. In day 150 and 180, the weights of their abdominal fat are 37.70 and 47.67 g, not significantly.The IMP content of hens' leg muscle increase significantly after day 120 while the IMP content of chest muscle have no significant difference during 4 stages. The IMP content of roosters' leg muscle decrease after the first rise, significantly, and the maximum(0.78mg/g) comes out in day 150.The IMF of chest muscle and leg muscle increase significantly with the growth of age before day 150 and stay constant later in hens. The P values of chest muscle leg muscle are 0.05 and 0.01. The IMF of leg muscle increase significantly(P<0.05) before day 120 while that of chest muscle has no change with age.The IMF content has no significant difference between hens and roosters in both chest muscle and leg muscle. IMF content of leg muscle comes out to be bigger than that of chest muscle in both hens and roosters, significantly(P<0.01).In hens, concentration of serum' VLDL becomes larger till day 180 and the maximum is 194.79mg/ml, while that of LDL and TG have no difference before day 120, and decrease in day 150, significantly. Concentration of HDL, TC, and FFA stay stable with age. Lipid parameters of roosters have no change with growth of age.1 Comparison in lipid levels between hens and roosters tell us:TC content of hens(2.75nmol/L) is less than that(3.27nmol/L) of roosters in day90.VLDL's concentration in hens are bigger than that of roosters after day 120, and TG content are bigger in day 150(0.91nmol/L, 0.30nmol/L)and 180(0.34nmol/L, 1.30nmol/L).Content of LDL in hens is less than that in roosters.IMF of leg muscle is negatively correlated with TC and LDL(R=-0.581,R=-0.433),and IMF of chest muscle is negatively correlated with LDL(R=-0.576) while that of TC is significantly(R=-0.448)3 Expression pattern of intramuscular fat metabolism candidate genes and correlation with IMFExpressions of Apo-B ? A-FABP grow up with age in chest muscle, significantly(P<0.01) till day 150 and day 120, respectively, when the maximum comes out.Expressions of PPAR-? and Apo-B decrease after the first rise in day 120 in hens' chest muscle, while that of PPAR-?, Apo-B and L-FABP decrease after the first rise in day 150 in hens' liver. Expression of Apo B increases till day 150. Expressions' up-regulation of FAS?Apo-Bin liver and that of Apo-B?A-FABP?PPAR-? in muscle can promote fat deposition in Wuhua chicken.The results show that VLDL, HDL, TG can be used for the breeding of Wuhua chickens' fat deposition. FAS, Apo-B in liver and Apo-B, A-FABP, PPAR-? in muscle are important genes in regulating the fat metabolism...
