The purpose of determination melamine residue in animal derived food(milk, eggs, chicken, pork), HPLC and GC/MS methods were established.Milk, eggs, chicken, pork were put to centrifugation tube, added treichoroacetic and lead acetate in turn, high speed blended, oscillation and centrifugation. supernatant was clean by MCX SPE, dried at 50? under a stream of nitrogen, dissolved in methanol, filtered 0.45mm. With the condition of Agilent C8 column(4.6*150mm,?5?m) detection by HPLC. The ampicillin extract was analyzed by HPLC with uv detection at wavelength of 240nm, mobile phase was acetonitrile/lOmM sodium heptanesulfonate+lOmM citrate buffer:8/92 (v/v).The calibration curve was showed a good linear relationship in rage of 0.05-50?g/g. the correlation coefficients are both more than 0.999,The recoveries of melamine added to sample were more than 80%,and the CV% within day was less than 7%, The between day CV% was less than 10%.the detection limits was 0.05?g/mL in milk,. 1?g/g in eggs,2?g/g in chicken and pork.The melamine analyzed in selective ion monitoring(SIM) mode using m/z 86,233,272 and 318 with electronic ionization (El) by GC/MS. The calibration curve was showed a good linear relationship in rage of 5-500ng/g. the correlation coefficients are both more than 0.999,The recoveries of melamine added to sample were more than 80%,and the CV% within day was less than 8%, The between day CV% was less than 10%.the detection limits was 30ng/g in milk,.40ng/g in eggs,50ng/g in chicken and pork..The result showed this method was sensitive and precisable to measure melamine residues in animal derived food.The established method will provide a new method for determination of melamine residues in animal derived food and monitor melamine residues in animal derived food to ensure and vindicate people healthness with academic and practicai references.
