This is the first detailed report of detection of these seven bee viruses in Apis mellifera and Apis cerana in Anhui province. It will help bee researchers and related institutions screening the present situation of honey bee health in Anhui, and warn them of the potential threat to sustainable development of apiculture.This paper includes two parts as below:1. Reprots on occurrences of honey bee virues in Anhui province; 2. System established of honey bee larvae incubation in vitro.In this study, using reverse transcriptase PCR and sequence analysis, a survey of seven honey bee viruses was conducted, covering almost the whole of Anhui province, China. Ten adult bees per tube were collected from apiaries in 21 towns, including 28 tubes of Apis mellifera and 10 of Apis cerana. The results showed as below:1. In the A. mellifera samples, frequencies were as follows:64% of apiaries were infected with DWV,43% with IAPV,32% with CBPV,14% with ABPV,11% with BQCV, and none with SBV or KBV. In A. cerana samples, the following four viruses were identified: DWV (80% of apiaries), IAPV (40%), CBPV (30%), and ABPV (10%). Simultaneous multiple viral infections were common in both species.2. Sequencing analysis show that DWV with Accession NO. JX878304.1 (97%); IAPV with NO. EU224279.1 (99%); ABPV with NO. HQ897161.1 (98%); BQCV with NO. GU825962.1 (95%); CBPV with Accession NO. KJ437447.1 (97%).3. DWV, IAPV, CBPV, ABPV and BQCV were infested almost in all zones in Anhui, excpeted SBV amd KBV.We hope this will help bee researchers and related institutions screening the present situation of honey bee health in Anhui, and warn them of the potential threat to sustainable development of apiculture.In the second part of this paper, an artificial in vitro culture system for honeybee was established.This assay part aims to establish an artificial in vitro culture system for honeybee, to provide theoretical basis and research conditions for study of honey bee virology indoor.We had honeybee pupae cultured successfully, the results showed that there was no significant difference between artificial in vitro bees and normal incubation ones.By feeding nutrition solution that contained DWV, larvae would be infected DWV easily, which wing would be deformed and life span declined greatgully, contrast with the CKs.
