Populus euphratica Oliv.(P.euphratica)is a origin of ancient relict plant with the characteristic of heterophylly,which is Belongs to Populus subgenus of Salicaceae.It's the constructive species and dominant species in arid desert region of Xinjiang,also the only tree species can naturally form a large area of forest in the arid desert region,and play an important role for the maintenance and regulation of fragile ecosystems of the arid desert regions.In this study,we studied the characteristic of the leaf morphology of P.euphratica and the distribution characteristics of endogenous hormones,and also analysised the expression profiles of different shaped leaves of different stages.The results showed that:(1)The heterophyll morphological features of P.euphratica are spatiotemporal characteristics with individual developmental stages.Displays in: 1)On tme,the leaf index and the length decreased gradually with the development of individuals with the individual developmental stages,and the leaf width,leaf area,petiole length,leaf margin serrate and leaf base angle is increased gradually,and has the remarkable difference between young and adult plants.2)On the space,leaf index and leaf length from the bottom to the top of the crown are gradually decreased with increasing height of P.euphratica at the same stage,and the leaf width,leaf area,petiole length,leaf margin serrate and leaf base angle is increased gradually,and there is a significant difference the top and bottom.(2)The endogenous hormones distribution characteristics of P.euphratica leaves: the trends of IAA and IAA / ABA ratio changes are decreases gradually with the individuals increasing ages and the crown level,and the trends of ZR,ABA,GA3 / IAA,ZR / IAA ratio changes are the opposite.The changes of IAA,ZR,ABA,IAA / ABA ratio,GA3 / IAA ratio and ZR / IAA ratio may have some connection with the change of leaf.(3)Analysis of expression profiling of P.euphratica leaves,the expression profiles of leaves in different developmental stages were sequenced.There are 602 differentially expressed genes between lanceolate leaves(Pe_B2)and strip leaves(Pe_B1)in B stage,which there are 333 up-regulated genes and269 down-regulated genes.In stage C,there are respectively 11272 and 9757 differentially expressed genes which ovate leaves(Pe_C3),lanceolate leaves(Pe_C2)relative to the strip leaves(Pe_C1),whichrespectively 7821 and 6438 up-regulated genes and 3451 and 3316 down-regulated genes.There are 115up-regulated genes and 376 down-regulated genes are the same differentially expressed genes in C stage.There are 2567,2655 and 2160 differentially expressed genes respectively in D stage broadly ovate leaves(Pe_D4),ovate leaves(Pe_D3),lanceolate leaves(Pe_D2)relative to the strip leaves(Pe_D1),which the number of up-regulated genes are 782,748,1084 and down-regulated genes were 1785,1907 1072 respectively.There are also 147 up-regulated genes and 408 down-regulated genes are the same differentially expressed genes in D stage.Compared with strip leaves(Pe_A1)in A stage,broadly ovate leaves in D stage,ovate leaves in C stage and lanceolate leaves in B stage are respectively have 4385,4564 and 1590 differentially expressed genes.And respectively consist of 2660,2915 and 888 up-regulated genes and 1725,1649 and 702 down-regulated genes,where there are 85 up-regulated genes and 221down-regulated genes are the same differentially expressed genes.For differential gene GO classification analysis,differences in gene function in a cell component mainly in cells and components,cellular organelles and other components at the molecular function mainly in catalytic activity,binding activity,transporter activity and molecular transduction activity,etc.function in biological processes is in the metabolic processes,cellular processes,stimulating the response of biological processes.KEGG metabolic pathway analysis showed that carbon metabolism,amino acid synthesis,photosynthesis,plant signal transduction,plant pathogen interactions,starch,sucrose metabolism,cysteine ? ? and methionine metabolism,amino sugars and nucleotide sugar metabolism,linolenic acid metabolism,flavonoid biosynthetic pathway is more enriched genes.(4)Analysis all differentially expressed genes showed that cytochrome P450,LG1(LIGULELESS1),FLC(FLOWERING LOCUS C)and other 11 genes related to leaf morphogenesis also may be involved in P.euphratica leaves formation.The response to stimulus is the most differentially expressed genes involved in biological processes after metabolic processes and cellular processes.And these genes metabolic pathways analysis also showed some genes of them involved in the cell enlargement and plant growth,such as involved in hormone transduction,etc.These genes may be a role for different P.euphratica leaves formation.But these genes need further validation.(5)Selected eight differentially expressed genes randomly,and using q RT-PCR method to detect them,the result showed that six genes expression patterns of them are consistent with expression profiling results.
