Fever is a common symptom of domestic animals,they often accompanied by pain,livestock growth was inhibited and performance was reduced by fever,due to heat stress,immunity was decreased,therefore,the use of antipyretic and analgesic drugs is more frequent.At present,the commonly used anti-inflammatory are injection by dipyrone and aminopyrine,but antipyretic effect of them has been less significant.Nimesulide is a drug for medical use who has better antipyretic effect.It is a non-steroidal anti-inflammatory drugs,more attention.At present,nimesulide tablets,granules has been used.Both dosage forms could cause gastrointestinal irritation and other adverse reactions,this has proved.Nimesulide although not apply to animals,but according to the actual situation of human health,the study of administration of transdermal formulation by the percutaneous route has applied value.Studies have shown that,in recent years,Radix volatile oil not only has a good antipyretic and analgesic effect,but also promote other medicines transdermal effect.Therefore,the study of traditional Chinese medicine which has the effect of antipyretic and analgesic is necessary.Based on the above analysis,this study intends to produce the compound film that can relieve fever and analgesia.Radix volatile oil were compared with the menthol in the effects of on transdermal penetration to nimesulide,screening the best concentration of penetration enhancers,on the basis of this jieguo,to observe the relieving fever and analgesia,analysis the pharmacokinetic characteristics of nimesulide in the compound film in different animals,provide a scientific basis for the preparation of compound film of volatile oil in radix bupleuri.Experiment ? Transdermal effect of volatile oil in radix bupleuri on nimesulide By comparing different concentration of volatile oil in radix bupleuri and menthol's transdermal effect on nimesulide which is heat analgesic drugs in the mouse skin,and the best transdermal and its concentration were screened.Intelligent instrument for percutaneous experiment was used,the mouse skin were choosed,and dose-effect relationship of different concentration of volatile oil in radix bupleuri on nimesulide's transdermal was researched,accumulated infiltration capacity in 12 hours and permeability coefficient were calculated.The result shows that accumulated infiltration capacity and permeability coefficient of nimesulide transmented by oil in radix bupleuri and menthol are both higher,accumulated infiltration capacity and permeability coefficient of nimesulide transmented by 8%oil in radix bupleuri is highest,higher than others extraordinarily.It shows that transdermal effect of 8%oil in radix bupleuri,according to this,the best promoting transparent program of nimesulide is determined.Experiment ? The production of the compound recipe film of volatile oil in radix bupleuri and its skin safety test Based on the determination of volatile oil in radix bupleuri as the film's transdermal,the best preparation of the film was screened,the film was producted,and its safety was initial evaluated.Polyvinylpyrrolidone K-30(PVP K-30),ethyl cellulose(EC)and triacetin(GTA)was choosed as the key index,L9(34)orthogonal experiment was designed,and comprehensive score of the film was estimated according to the film's nature and appearance,and the best prescription was screened.This result shows that 2g of PVP K-30,1 g of EC,5mL of GTA is the best prescription,and the film produced by this membrane has both property and safety.Experiment ? Effect of relieving fever and analgesia of the compound recipe film of volatile oil in radix bupleuri Effect of relieving fever and analgesia of two films of relieving fever and analgesia were researched,film 1 was compound recipe film of volatile oil in radix bupleuri,film 2 was recipe film of volatile oil in radix bupleuri.Animal fever model was established,rabbits were randomly divided to 5 groups,two films were administrated through skin respectively,and body temperature of rabbits was measured 5 hours after medication;mouse that fit the basic pain threshold screened by Hot-plate were randomly divided to 5 groups after poodled.The heat test shows that both of film 1 and film 2 have better effect on relieving fever,the speed of relieving fever of film 1 is faster than film 2;Analgesia test shows that both of film 1 and film 2 have better effect on analgesia,film 1 is faster than film 2.The result shows that,the effect of relieving fever and analgesia of compound recipe film of volatile oil in radix bupleuri is better,the effect of joint application of volatile oil in radix bupleuri and nimesulide is better than volatile oil in radix bupleuri used individually.Experiment ? Nimesulide's pharmacokinetics determination in animals Pharmacokinetics of nimesulide in compound recipe film of volatile oil in radix bupleuri was researched in rabbits and pigs.After the rabbits and pigs were painted,blood drug level of nimesulide in 48 hours was determined by high-performance liquid chromatography quadrupole tandom mass spectrometer(LC-MS/MS)in animals,pharmacokinetics parameters were analysed by winnonlin 6.2.0 noncompartmental model.The result shows that,the peak of blood drug level in rabbits is 61.9?g/mL,peak time is 12.4h,drug elimination half life of elimination half life is 40h,the peak of blood drug level in pigs is 528.00ng/mL,peak time is 4h,drug elimination half life of elimination half life is 19.63h,The test result shows that,nimesulide in the film can penetrate skin to blood,and maintain unvarying blood drug level.
