Since genetically modified crops commercially planted in1996, transgenic crops and its industry dramatically developed, at the same time the safety of GMO products has been widely questioned. GMO undergo rigorous safety assessment before being allowed into the market on a large-scale, allergenicity assessment is one of important aspect. First, we genome-widely screened and characterized rice endogenous allergens by full-length amino acid sequence alignment. Then, in order to study the unintended effect possibly caused by insertion of exogenous gene, transgenic CrylAb/Ac rice Huahui1(HH1) and its non-transgenic counterpart Minghui63(MH63) were selected as research objects. Conventional two-dimensional electrophoresis was taken to compare the total proteins of the mature seeds. Finally, based on2-DE and immunoblotting, serum screening was performed for rice seeds endogenous allergen. The main results were as follows:(1)122putative rice allergens were identified and characterized by sequence alignment between rice proteins and known allergens, including the22rice allergens in present databases. Some allergens proteins presented as a multigene family and clustered on the rice chromosomes;(2) Modified phenol extraction method is more suitable for the extraction of rice seeds proteins than TCA-acetone method.(3)77protein spots from2-DE map of total rice seeds proteins were successfully identified, which possess catalytic activity, binding, nutrient reservoir activity, etc., and involve in metabolic process, single organism process, celluar process, etc.(4)7potential rice allergens were discovered among the77identified rice proteins.
