Objective Through the effects of jujuboside on the the expression of ingestioncontrol gene in hypothalamus primary cell,explore the function method of jujuboside adjusting the feed intake.Methods The chicken embryos eggs needed in the experiment provided by the Liaoning Medical University hatching room.The test planed to select the younger chicken embryos, taken the hypothalamus to cultivate the hypothalamic primary neurons cells, and application of cellular immune histochemical method and immune protein imprinting technique were used to identify the cultured cells into neurons.Selection of well developed cells, respectively in the medium, adding different concentration of jujuboside, HSP70 and CRH as processing factor and added different concentration of jujuboside, HSP70 and CRH as processing factor and raise the temperature of 40.5 to ?simulate the thermal stress, adopt the method of Real- time PCR and Western Blot analysis of hypothalamic NPY? Ag Rp?GABA A-?1?MC4R?POMC?HSP70 and CRH m RNA expression, using SPSS statistical analysis software, clarify the stress on the effect of feed intake adjusting factor and jujuboside of its adjustment.Results1.Successfully cultivate chicken hypothalamic neurons primary cells, and identify the cells as develop the hypothalamus neurons. 2.Compared with normal control group, group of jujuboside added the NPY?GABA A-?1?MC4R?Ag Rp significantly increased(P< 0.01).POMC and CRH m RNA expression were significantly lower(P < 0.01). 3.The group of HSP70: NPY and GABA A-?1 m RNA were significantly higher than that of normal control group(P < 0.01), MC4 R and CRH were significantly lower than normal control group(P< 0.01), Ag Rp and POMC m RNA significantly lower(P< 0.01). 4.The group of CRH: NPY ? MC4 R ? POMC and CRH m RNA were significantly higher than that of normal control group(P < 0.01), GABA A-?1 is higher(P<0.05), Ag Rp were significantly lower(P< 0.01).The tendency is the same of the heat stress group(P>0.05). 5.Group of jujuboside:NPY?POMC and GABA A-?1 were significantly lower(P<0.01), increased the MC4R? Ag Rp?HSP70 m RNA expression(P<0.01). 6.The group of HSP70 during 40.5 ?.:NPY and POMC were significantly lower(P <0.01). GABA A-?1 is higher(P<0.05). The tendency of Ag Rp and MC4 R m RNA were the same of the heat stress group(P>0.05). 7.At 39.5?, Correlation analysis showed that the NPY m RNA is positively related to the POMC and HSP70 m RNA(r2=0.777, P=0.020;r2=0.806, P=0.015);At 40.0?,MC4 R and POMC m RNA negatively correlation(r2=0.840,P=0.029).Conclusions 1.Successfully culture chicken primitive cells in the hypothalamus, and identify the cultured cells into neurons. 2.The group of Jujuboside and HSP70 promote could relate genes in the neurons of broiler chicken hypothalamus.3.The group of CRH and 40.5 ?of heat stress, could relate genes in the neurons of broiler chicken hypothalamus.And the NPY?POMC?GABA A-?1?MC4R?Ag Rp and CRH were in the same tendency.CRH could Instead of heat stress group by experimental conditions. 4.The group of Jujuboside and HSP70 promote could restore genes of NPY?POMC?GABA A-?1?MC4R?Ag Rp and CRH in the neurons of broiler chicken hypothalamus during 40.5 ?. 5.The NPY m RNA is positively related to the POMC and HSP70 m RNA,MC4 R and POMC m RNA negatively correlation.
