The image acquisition of flow cell using microfluidic chip and lensless CMOS image sensor is a hot-spot in recent years.However,due to the lack of lens amplification,the collected images have low resolution and less detailed information And the control effect of microfluidic is also not ideal,leading to the cell adhesion and ghosting phenomenon,therefore,the study of cell image acquisition in flow cell detecting is of great significance to cell analysis and detection.In this paper,the microfluidic technology and CMOS image sensing technology are used to study the flow cell imaging,the main contents are: the design of microfluidic chip with three dimensional focusing function, the analysis and improving of the chip's surface roughness, the study of technology that use image sensor and PDMS to fabricate the microfluidic chip,the hardware implementation of cell image acquisition and the preprocessing of raw cell images.Firstly, the focusing principle and design process of microfluidic chip are studied, and the structural characteristics and defects of trigeminal microfluidic chip with two-dimensional focusing performance are summarized and analyzed. On this basis, the rigeminal microfluidic chip with a baffle in middle channel, which could complete the cell focusing in the vertical direction, is fabricated. Secondly, explore the fabrication process of microfluidic chip,analyze the key factor that affect the roughness of the chip's inner wall and propose the solution. Then,study the process flow that use the glass cover of CMOS sensor, instead of the traditional glass,to combined with PDMS, The microfluidic chip based on OV5640 image sensor is manufactured. Finally, complete the hardware implementation of cell image acquisition,storage and display. And preprocess the original cell image to reduce the amount of data processing.Through the research of the paper, the focusing performance and control effect of the microfluidic chip are improved, and a cell image acquisition system based on FPGA hardware platform is constructed to contribute to further reducing the system volume and improving the processing speed.
