| Iron deficiency anemia is a global nutritional deficiency disease caused by the long-term lack of iron and has a serious impact on human health.Iron supplementation is the most effective way to treat iron deficiency anemia.The common iron supplements are ferrous iron supplements,but they have a stimulating effect on the gastric and intestinal mucosa.Therefore,it is of great significance for the research and development of new iron supplements.As the third generation iron supplement,polysaccharide iron has the advantages of high iron content,good stability,high bioavailability and small side effects,.And it has become a research hotspot at home and abroad.In this study,corn starch syrup and ferric chloride hexahydrate were used as raw materials to prepare corn starch polysaccharide iron.Single factor experiments and response surface experiments were used to investigate the complex reaction of six hydrated ferric chloride with syrup and various factors of alcohol precipitation and washing process.It is able to optimize the operation process to realize the mass production of iron polysaccharide.The raw material ferric chloride hexahydrate quality testing,including the content,heavy metal and arsenic salt detection.The test results showed that the content of heavy metals did not exceed 5ppm;the content of arsenic did not exceed2 ppm.The content of ethanol in the product was determined by gas chromatography.The methodology experimental results showed that the methods were accurate and reliable,and could meet the need of analysis.The parameters of the synthesis process were optimized.The reaction temperature,time,system p H,material ratio and stirring rate were optimized by single factor experiment and response surface optimization.The optimal process parameters were determined as reaction temperature 73.6℃,reaction time 3.8h,reaction systemp H11.02,stirring rate 90 r / min.The product iron content is 44.58% under the best process parameters.The parameters of alcohol precipitation,drying process were optimized,95%ethanol was used to make alcohol precipitation of the reaction solution.Through the single factor experiment and response surface optimization,the proportion of ethanol,the speed of centrifugation,the centrifugation time,the re-dissolved water ratio,the drying time were investigated.The optimum process parameters were determined as follows: ethanol ratio 76.5%,centrifugal speed 5830 r / min,centrifugation time 20 min,the ratio of th re-dissolved water 8: 1,drying time 6h.Under the process parameters,the iron yield was 52.53% and the ethanol residue was 0.26%.The products were identified and analyzed by chemical methods(iron salt identification,anthrone identification)and spectroscopy(UV,IR,HPLC).The results showed that the structure of corn starch polysaccharide iron was similar to the original drug. |
