Capillary electrophoresis is widely used in the separation of amino acids,monosaccharides and chiral drugs,due to its advantages of high resolution,low consumption and easy operation.In this study,capillary electrophoresis was used to investigate the separation of amino acids in vinegar,amino acids and monosaccharides in Traditional Chinese Medicines,and enantiomers of propranolol.The established methods were validated.1)In this study,the capillary electrophoresis method with ultraviolet detection was established to determine underivatized amino acids in vinegar,according to the coordination interaction between amino acids and copper ions.The online sweeping technique was combined to improve the detection sensitivity.The quality of vinegar was evaluated with amino acids as parameters by United Nations Food Agriculture Organization/Word Health Organization amino acids model and principal component analysis.The factors affecting the separation of amino acids,such as concentration of copper sulfate,pH of background electrolyte,capillary diameter,separation voltage and injection time were optimized by single rotation method.Optimum conditions were obtained:under 50 mmol/L CuSO4?adjusting p H to 4.40?as running buffer,70s injection time,22.5 kV separation voltage and and 254 nm detected wavelength.As a result,ratios and comprehensive scores of amino acids in sample S11 were most close to stipulates and the highest.2)The micellar electrokinetic capillary chromatography method was established to simultaneously separate 16 amino acids and 6 monosaccharides in Traditional Chinese Medicines.Sodium dodecyl sulfate was used as surfactant.The proposed method was applied to the analysis of Lycium barbarum and Chinese Angelica.The effects of separation mode,type and concentration of surfactant,concentration of borate solution,pH of background electrolyte and separation voltage on the simultaneous separation of amino acids and monosaccharides were systematically investigated.The possible separation mechanism was also discussed.Optimized separation conditions:40 mM borate solution and 40 mM sodium dodecyl sulfate;pH was 10.02;the separation voltage was 11 kV in the first 20 min and 13 kV after 20 min;the detection wavelength was 266 nm.Under the established conditions,effective separation of 16 amino acids and 6 monosaccharides were achieved within 30 min.3)A mixed micellar electrokinetic capillary chromatography was established for the separation of propranolol enantiomers.Chiral surfactant?Sodium deoxycholate?was used as chiral selector,and nonionic surfactant?Polyoxyethylene lauryl ether?was used to form mixed micelles with sodium deoxycholate.The concentration of borate solution,the concentration of sodium deoxycholate,the concentration of polyoxyethylene lauryl ether,pH of background electrolyte and the separation voltage were investigated by single rotation.Separation conditions were as follows:30mM borate solution,15mM sodium deoxycholate and 30mm polyoxyethylene lauryl ether?adjusting pH to 9.31?was used background electolyte,the applied voltage was 15 kV and detection wavelength was 215 nm.Under optimal conditions,the effective separation of propranolol enantiomer was achieved within 12 min.
