The outbreak of algal blooms seriously impact aquatic environments,and threaten the health of aquatic animals and human beings.Compared with the physical and chemical methods,microbial methods for algae control is simpler.Currently,a new method for inhibition the growth of algal cells was invented,which is superior to viruses,bacteria and previous fungal methods.This fungus can degrade all of the algal cells in the fungus-alga co-cultured system.However,the mechanism of this new method is still unclear.In this study,based on the previous transcriptome and semi-quantitative PCR analysis results of T.versicolor-F21 a co-cultured with cyanobacteria of our team,real-time PCR and proteomic techniques were used to further verification previous results.The proteomic data was further compared with the results of previous transcriptome analysis to find the possible key decomposition enzymes and pathways which were further verified by experiments.The main results of the study were as follows:(1)Quantitative PCR result showed that Gene ID of 60067(GH16),135205(GH5)and 30530(GH13)might play an important role in degradation,which was consistent with the conclusion of previous results of transcriptome analysis.(2)Proteomic analysis of fungal mycelia samples co-culture with cyanobacteria at 0 h and 12 h shows that a total of 3,754 proteins were identified.Of those proteins,2,809 proteins could be quantified.205 proteins were significantly up-regulated,while 145 proteins were significantly down-regulated.(3)Compared with the results of quantitative proteomic analysis and transcriptome sequencing analysis,we observed that 3,319 transcripts or proteins simultaneously in both results.72 genes were simultaneously up-regulated in the results of the two analyzing methods.Pathways related to amino acid metabolism and membrane metabolism might play important roles in the fungal algicidal process as revealed by enrichment results both of proteomic data and transcriptome data.(4)Members of glycoside hydrolases,such as cellulases were highly expressed during the algicidal process.The results of further experiments showed that all the algal cells could be completely eliminated by cellulases within different times,while the degradation results of trypsase on algal cells was less effective.Combination of different results indicated that cellulases may play a major role in the elimination of cyanobacterial cells.
