| The active ingredient of the green peel of Juglans mandshurica was extracted with 95% ethanol,and then extracted with petroleum ether,chloroform and ethyl acetate.The extraction phase solution was separated by the chromatography and the GC-MS,and the active components were analyzed by the gas chromatography and the GC-MS.The petroleum ether extraction phase of petroleum ether acetate system was divided into nine substances,when the flow ratio of 3:7.While seven parts of the material can be separated at the ratio of 1:1,and six substances at 1:5.The fraction of the methanol-chloroform system of the ethyl acetate extraction phase separated eight substances when the ratio is 1:11.Based on the single factor test,an orthogonal design was used to optimize the extraction process of procyanidins from the exocarp of Juglans mandshurica.The results were as follows: the ratio of liquid to the material was 1:30,and the volume fraction of ethanol was 50%,the ultrasonic temperature was 70 ℃and the ultrasonic time was 30 min.As for the leaves,the optimal conditions were as follows: the ratio of liquid to the material was 1:25,and the volume fraction of ethanol was 50%,The ultrasonic temperature was 70 and the ultrasonic time was 50 mi℃ n.The effect factors for the test results are listed in descending order: the ultrasonic temperature,the ethanol volume fraction,the ultrasonic time and the liquid to the material ratio.The Bacteriostasis of Clostridium from the volume fraction of 95% ethanol extraction of the green peel of Juglans mandshurica extract,the chloroform extraction phase,the ethyl acetate extraction and the aqueous phase and the water solution 60% ethanol extraction of the green peel of the Juglans mandshurica extract were studied by the oxford cup method.There was an obvious antibacterial activity against the Gloeophyllum trabeum by the Chloroform phase solution from the 95% ethanol extracted Juglans mandshurica and the water solution of the 60% ethanol extracted Juglans mandshurica.And the petroleum ether phase and aqueous phase solutions extracted by 95% ethanol had obvious antibacterial activity against the coriolus versicolor.The total alkaloid of green peel and leaves of Juglans mandshurica was extracted by the ultrasonic method.The extraction process of total alkaloids was optimized by the orthogonal experiment method.And explore the antioxidant properties of total alkaloids.L9(34),the optimum conditions for the extraction of green peel of Juglans mandshurica were as follows: the ratio of liquid to material was 1:10,the volume fraction of ethanol was 80%,the ultrasonic temperature was 40 ℃,the ultrasonic time was 40 min.The optimum conditions for the extraction of leaves were as follows: the ratio of liquid to material was 1:15,the volume fraction of ethanol was 60%,the ultrasonic temperature was 50 ℃,and the ultrasonic time was 50 min.The results showed that the antioxidant capacity of the total alkaloid solution of the green peel from Juglans mandshurica has more effect on hydroxyl radical than those of superoxide anion and nitrite ions.The total alkaloid of the leaves from Juglans mandshurica had more effect on nitrite ion than hydroxyl radical and superoxide anion. |
