Korean pine nut is a seed of Pinus koraiensis Sivb.ct Zucc,which is rich in nutrients and has high oil content.Pine nut oil is rich in unsaturated fatty acids,including linolenic acid,arachidonic acid,pinolenic acid,with lipid-lowering,anti-oxidation,inhibition of appetite and other effects.However,due to the liquid properties of pine seed oil,and its easily oxidized and deteriorate in processing and storage,and limited in its application.Microencapsulation can protect pine nut oil from affecting by the environmental factors,such as oxygen,effectively improve its oxidation stability and application value.In this thesis,pine nut oil was extracted by aqueous enzymatic method,preparation of microcapsules by complex coacervation,the stability of pine nut oil and microcapsule and under simulated conditions of gastrointestinal test to evaluate the quality of microcapsule.The results show that:The optimal conditions of extract pine nut oil.The optimal enzyme preparation for the extraction of pine nut oil was selected as the measure of the extraction rate.Through single-factor and response surface methodology to investigate the extraction of pine nut oil.The optimal extraction conditions were obtained as follows:with ultrasound treatment at ultrasonic power 0.28 W/cm~2,50?for 40 min,4.08 h of hydrolysis using Alcalase protease at a dosage of1427 U/g with a substrate to water ratio of 1:5(g/mL)at 44?.Under the optimal conditions,the yield of pine nut oil reached 73.01%.The optimal conditions of preparing pine nut oil microcapsule.Pine nut oil microcapsules were successfully prepared using gelatin and arabic gum as wall materials by complex coacervation.The influences of the ratio of two wall materials(V-gelation:V-arabic gum),the mass ratio of core material,the concentration of wall materials and agglomeration time were determined.The optimal formulation were determined by orthogonal test.Meanwhile the physical and chemical properties and the effects of temperature on the oxidative stability of pine nut oil in microcapsules were also analyzed.The optimum preparation conditions of pine nut oil capsules were achieved when the ratio of two wall materials was 2:1,core/wall ratio 2:3,concentration of wall materials was 2%,agglomeration time was 50min.The encapsulation efficiency was 87.23%.The feature of the microcapsule was investigated.The moisture content,solubility of pine nut oil microcapsules was 5.1%and 98.09%.The interaction between gelatin and arabic gum was analyzed by IR spectroscopy(FT-IR),field emission scanning electron microscopy(FESEM).DSC analysis indicated that complete structure of microcapsules can exist stably during room temperature.The simulated conditions of gastrointestinal test showed that microcapsules have a good sustained release and enteric.The oxidative stability of pine nut oil and microcapsule.The results showed that the effects of temperature and light on the oxidation stability of pine nut oil were significant.Storage under 3?without light could markedly improve the stability of the pine nut oil and the microcapsulates.The oxidation of encapsulated pine nut oil revealed that microcapsules can enhance the storage stability,prolong the storage period of pine nut oil.
