| Euphorbia lathyris L is a kind of toxic traditional Chinese medicine(TCR),and has been used to remedy diarrhea,water,and hemorrhagic fever for a long time.Recent studies show that it contains fatty oils,diterpenoids,flavonoids,as well as a small amount of coumarin and volatile oils.However,previous studies are just limited the discovery the chemical components of it or the remedy of single component.It is of importance to study the synergistic effect and bioactivity systematically.Hence below works were performed in this thesis.1)Screening bioactive components against Hela cell using cell metabolomicsWith ultrasonic extraction method,ethanol total extract was obtained.Then it was separated by silica gel column.And totally four fractions(QJZ-E1,2,3,and 4)was achieved based on the polarity.Under optimum conditions,the cytostatic activities against Hela cell was investigated.Six extracts of Euphorbia lathyris L were used including QJZ-E and QJZ-E14.It was found that the cytostatic activity of QJZ-E3 was strongest among them.It indicates that the main active component against cervical cancer is in QJZ-E3.And then cell metabolomics was used to screen bioactive components against Hela cell.UPLC-MS resulted data was analyzed by principal component analysis(PCA)and partial least squares(PLS-DA),respectively.Besides Euphorbia Factor L1 and Euphorbia Factor L3 reproted before,ten more components were determined,including:deoxy Euphorbia Factor L1、Euphorbia Factor L8、Euphorbia Factor L15、Euphorbia Factor L16 and Euphorbia Factor L25.2)Antioxidant activity of different extracted fractions from Euphorbia lathyris LWith ultrasonic extraction method,ethanol total extract was obtained.Then it was separated by silica gel column.And totally four fractions(QJZ-E1,2,3,and 4)was achieved based on the polarity.And above anti-oxidized activities of four fractions and the totoal extrat were investigated by DPPH free radicals and ABTS+methods,respectively,with vitamin C and butylated hydroxytoluene(BHT)as controls.It was found that the scavenging effect of QJZ-E3 was the highest among five experiments.Furthermore,Ultra high performance liquid chromatography-mass spectrometry(UPLC-MS)was done to analyze the chemical components in QJZ-E3.And totally eight components were determined including:Aesculetin,Artemetin,Aurantiamide acetate,Quercetin-3-glucuronide,Aucosterol,Kaempferol-3-glucuronide,Vitexicarpin and Euphorbetin.3)Fabrication cerium phosphate chemical modified electrode for rapid and simultaneous detection of hydroquinone and its isomersHere one novel chemical modified electrode(CME)was prepared for determine the organic pollutants in water to quarantine the TCM quality from the source.It was fabricated by pasting cerium phosphate nanotubes onto a glassy carbon electrode(GCE)with nafion matrix.The morphologies and components of the coating were characterized by transmission electron microscopy,scanning electron microscopy and energy-dispersive spectroscopy.Cyclic voltammetry and differential pulse voltammetry(DPV)showed the specific surface of the electrode to be significantly increased and the electron transfer rate to be accelerated.The modified GCE was applied to the determination of hydroquinone(HQ)via DPV.The oxidation current increases linearly in the 0.23μM to 16 mM HQ concentration range which is as wide as five orders of magnitude.The limit of detection is 0.12μM(based on a signal-to-noise ratio of 3),and the sensitivity is 1.41μAμM-1 cm-2.The method was further applied to the simultaneous determination of HQ,catechol and resorcinol.The potentials for the three species are well separated(20,134,and 572 mV vs SCE).Average recoveries from(spiked)real water samples are between 95.2 and 107.0%,with relative standard deviations of 0.9-2.7%(for n=3)at three spiking levels.The method was validated by independent assays using HPLC. |
