Antrodia camphorata is a medicinal fungus native to Taiwan.Modern medical research shows that its extract has anti-tumor,antiviral,anti-inflammatory,detoxification,antioxidant and hepatoprotective activity.Its active ingredients are mainly three terpenoids and polysaccharides.The results showed that the three terpenoids in the plant had anti-tumor,detoxication and liver protective activity.Wild camphor grows slowly and yields very little.Firstly,the liquid fermentation process of Antrodia camphorata nut was systematically studied.In this paper,single factor experiments and orthogonal experiments were combined to optimize the liquid fermentation medium.In the deep liquid fermentation medium,the factors affecting the mycelium and total polysaccharide content and the content of three terpenes were glucose,cornmeal and bran.The liquid culture conditions,such as culture temperature,time,rotate speed,volume of liquid and so on,were studied.The optimum fermentation conditions for liquid fermentation were 14 days,pH 4,dissolved oxygen 20%,temperature 28,corn surface as carbon source,bran as organic nitrogen source,and inorganic salt as magnesium ion.According to the orthogonal experiment,the optimum combination of A3B3C2D1 and MgSO4 was 0.15%,which was 0.4%for wheat bran,3%for corn flour and 0.25%for KH2PO4.In this paper,the effective constituents in mycelia of Antrodia camphorataa were extracted by enzymolysis and ultrasonic method.The results show that the optimum extraction conditions of Antrodia camphorata terpene for enzymolysis time was 2h,pH 5.5,2%cellulase dosage of polysaccharides;the optimum extraction conditions of Antrodia camphorata were solid-liquid ratio(m/V)1:30,ultrasonic power 120W,extraction time 35min,extraction temperature of 55DEG C.In this paper,we studied the effect of Antrodia camphorata extract on proliferation of HepG2cells.The results showed that the proliferation of HepG2 cells could be inhibited by Antrodia camphorata extract at concentration 12.5-400 g/mL significantly..In this paper,the preparation and formula of the oral liquid of Antrodia camphorata were discussed.According to Antrodia camphorata extract 2%,fructose syrup 4%,beta cyclodextrin0.35%,xylooligosaccharides 2%,CMC-Na 0.10%,xanthan gum 0.04%,sodium alginate 0.06%deployment,regulated by NaHCO3 pH 7 to 8.0,121℃sterilization 10min,Antrodia obtained oral liquid.In summary,in this study,deep liquid fermentation technology was used to produce the mycelia of Antrodia camphorata which contained three terpenoids.Meanwhile,the extraction methods of two functional components of three terpenoids and polysaccharides were studied.On this basis,the human hepatoma cell HepG2 was used as a model to detect the effect of extracts on the proliferation of tumor cells.Finally,the oral liquid of anti-tumor was prepared with the extract of Antrodia camphorata extract as the main raw material.
